Transcriptomic studies of mouse naïve and Th17 cell from wildtype or Malat1 deficient background

Project description:Long non-coding RNAs (lncRNAs) are critical regulators of mammalian gene programs. Metastasis Associated Lung Adenocarcinoma Transcript 1 (Malat1) is the most abundant lncRNA expressed in intestinal Th17 cells critical for maintaining tissue homeostasis and regulating local inflammation. Here, we report that Malat1 negatively regulates IL-17A and IL-17F productions in intestinal Th17 cells during colitis and contributes to local inflammation. Global RNA interactions with DNA by deep sequencing (GRID-seq) coupled with transcriptomic studies revealed Malat1 is recruited to the Il17a-Il17f super enhancer in Th17 cells, and regulates Il17a-Il17f transcription.

Effect of HDAC1 deficiency on in vitro Th2 and pathogenic Th2 differentiated murine CD4+ T cells

Project description:The differentiation of naïve CD4+ T cells into distinct effector T helper (Th) subsets is controlled by the chromatin state of the cells which in turn is regulated by various epigenetic mechanisms such as reversible lysine acetylation mediated by histone acetyltransferases and histone deacetylases (HDACs). In addition, Th2 cells can aquire pathogenic features, secreting pro-inflammatory factors and high levels of the cytokine IL-5. By using HDAC1 deficient naïve CD4+ T cells and establishing a protocol for differentiation of pathogenic Th2 cells, we could establish a critical role for HDAC1 in regulating the differentiation and pathogenicity of in vitro generated pathogenic Th2 cells

Project description:The goal of this assay is to investigate the signaling pathways involved in dopamine's promotion on Th2 differentiation and proliferation and to clarify the role of dopaminergic nerve innervation in neonatal asthma pathogenesis, to help identifying potential targets for therapy. To do that, we isolated naïve CD4+ T cells from spleens of non-transgenic C57BL/6 mice according to established protocols, and we cultured them under Th0 condition with/without dopamine treatment. To obtain an unbiased view of gene regulation in mouse CD4+ T cells exposed to dopamine, we analyzed the transcriptome of untreated control CD4+ T cells ('Th0') and dopamine-treated CD4+ T cells ('Th0+Dopa') via explorative RNA-sequencing.

Project description:Previously, lncRNA Malat1 knockout mice were generated by insertional inactivation. By crossing this line to MMTV-PyMT mammary tumor mouse model, we produced PyMT;Malat1 wild-type (WT) and PyMT;Malat1 knockout (KO). Furthermore, we generated Malat1 transgenic mice by targeting ROSA26 locus and bred them to PyMT;Malat1 knockout mice to produce Malat1-rescued PyMT;Malat1 knockout;Malat1 transgenic animals (TG). Using mammary tumors from the three groups of animals, we performed RNA-Seq analysis to identify differentially up-regulated genes in KO tumors to find novel target genes of YAP-TEAD pathway.
