Human A549 and MRC5 Cell Response to HCoV-229E Infection

ABSTRACT: The purpose of this experiment was to evaluate the human host cellular response to wild-type Human coronavirus strain 229E (HCoV-229E) infection. Sample data was obtained for mock and infected (MOI 5) immortalized human lung epithelial cells (A549) and immortalized human lung fibroblasts cells (MRC5) processed for RNA sequencing (RNA-Seq) expression analysis.

Transcriptomic profiling of human corona virus (HCoV)-229E -infected human cells

Project description:Human coronaviruses (HCoVs) cause mild to severe respiratory infection. Most of the common cold illnesses are caused by one of four HCoVs, namely HCoV-229E, HCoV-NL63, HCoV-HKU1 and HCoV-OC43. Several studies have applied global transcriptomic methods to understand host responses to HCoV infection, with most studies focusing on the pandemic severe acute respiratory syndrome coronavirus (SARS-CoV), Middle East respiratory syndrome CoV (MERS-CoV) and the newly emerging SARS-CoV-2. In this study, Next Generation Sequencing was used to gain new insights into cellular transcriptomic changes elicited by alphacoronavirus HCoV-229E. HCoV-229E-infected MRC5 cells showed marked downregulation of superpathway of cholesterol biosynthesis and eIF2 signaling pathways. Moreover, upregulation of cyclins, cell cycle control of chromosomal replication, and role of BRCA1 in DNA damage response, alongside downregulation of the cell cycle G1/S checkpoint, suggest that HCoV-229E favors S phase for viral infection. Intriguingly, more than 80% of key factors of cell innate immunity, interferon-stimulated genes (ISGs) and other transcripts of early antiviral response genes were downregulated early in HCoV-229E infection. This study will enhance our understanding of commonly circulating HCoVs and hopefully provide critical information about still-emerging coronaviruses.

Immune factors produced by PBMCs upon stimulation with Lactobacillus delbrueckii ssp. bulgaricus OLL1073R-1-derived exopolysaccharides inhibit HCoV-229E and SARS-CoV-2 replication

Project description:The COVID-19 pandemic entered a controllable phase due to vaccine and drug development. The development of defense strategies of enhancing host innate immunity remains imperative to counter the emergence of infectious SARS-CoV-2 variants. In this study, we examined the inhibitory effects of Lactobacillus delbrueckii ssp. bulgaricus OLL1073R-1 (OLL1073R-1)-derived exopolysaccharides (R-1 EPS), a natural immunomodulator, against coronaviral infection. The inhibitory effect of R-1 EPS on HCoV-229E was evaluated first and subsequently extended to SARS-CoV-2. Cell culture supernatant was obtained by stimulating human peripheral blood mononuclear cells with R-1 EPS (R-1 sup), and the inhibitory effect of R-1 sup on coronaviruses was evaluated using the human lung cell line, MRC5. High levels of proinflammatory cytokines were detected in R-1 sup, and replication of both HCoV-229E and SARS-CoV-2 was significantly suppressed in MRC5 cells pretreated with R-1 sup. Transcriptomic analysis revealed that type I IFN signaling was upregulated in R-1 sup-treated MRC5 cells. Our results suggest that immune factors, including proinflammatory cytokines, in R-1 sup, enhance type I IFN response, resulting in a broad spectral inhibitory effect on coronaviruses. This study provides a scientific basis for the application of OLL1073R-1 and R-1 EPS as preventive strategies against SARS-CoV-2 and other coronaviruses.

All experiments have been performed at 33°C which is required for sufficient viral entry/replication. Also included are two samples which have been treated with a heat inactivated coronavirus (56°C for 5min).

Characterizing immune signatures of seasonal coronaviruses HCoV-229E and -OC43 in human nasal airway epithelial cells

Project description:Seasonal coronaviruses, including HCoV-229E, -NL63, -OC43, and -HKU1, are prevalent worldwide, predominantly causing mild, self-limiting upper respiratory (re-)infections in adults, often presenting as the common cold. However, in individuals with compromised immune systems, these viruses may lead to more severe illness and even fatalities. Recently, there has been a renewed interest in studying HCoVs due to their amenability to handling in reduced biosafety containment, offering valuable alternatives to SARS-CoV-2 for preclinical screening and the development of antiviral treatments. Despite their significance, research on HCoVs has been hindered by limited host-genomic data. To address this, we performed RNA-sequencing on 3D air-liquid interface human nasal airway epithelial cells (hNECs) infected with the alphacoronavirus HCoV-229E and the betacoronavirus HCoV-OC43. These hNECs were derived from pooled adult donors and exhibited pseudostratified mucociliated differentiation, faithfully replicating the complexities of normal airway biology. Our study aimed to identify specific immune signatures associated with HCoV infections in a physiologically relevant model. By elucidating the host responses induced by different seasonal coronaviruses, we can gain valuable insights into their pathogenesis and interactions with the respiratory epithelium. This knowledge may pave the way for the development of targeted therapeutics and prophylactics to combat HCoV infections effectively.
