Project description:Peripheral artery disease (PAD) can cause limb pain and ulcers that won't heal. It is a leading cause of amputation, yet the moleculer mechanims that drive disease are poorly understood. To gain insights into mechanims of PAD at the transcriptome level, we performed sc-RNA-seq on non-ischemic and ischemic tissue from a male PAD patient who had undergone a below knee amputation.

Suppression of ischemia in arterial occlusive disease by JNK-promoted native collateral artery development

Project description:Arterial occlusive diseases are major causes of morbidity and mortality. Blood flow to the affected tissue must be restored quickly if viability and function are to be preserved. Collaterals are artery-to-artery or arteriole-to-arteriole interconnections that can bypass an occlusion by providing an alternative route for blood flow to the affected tissue. The increased flow and sheer stress initiate processes that result in the remodeling (arteriogenesis) of these vessels into efficient conductance arteries. Here we report that the mixed-lineage kinase (MLK) pathway activates cJun NH2-terminal kinase (JNK) in endothelial cells. Disruption of Mlk2/3 or Jnk1/2 genes caused severe blockade of blood flow and failure to recover in the femoral artery ligation model of hindlimb ischemia because of abnormal collateral arteries. We show that the MLK-JNK pathway is essential for patterning and maturation of collateral arteries during development, but this pathway is not required for angiogenesis or arteriogenesis in adults. JNK in endothelial cells promotes Delta-like 4-induced Notch signaling and suppresses excessive sprouting angiogenesis during development. This function of the MLK-JNK pathway contributes to normal formation of native collateral arteries. The MLK-JNK pathway is therefore a key regulatory mechanism for vascular development. These data highlight the crucial importance of the collateral circulation in the response to arterial occlusive diseases. RNA-seq analysis of mouse lung endothelial cells (MLEC) of the following genotypes Cdh5-Cre+ Jnk1+/+ Jnk2+/+ Jnk3-/-(ECtrl), Cdh5-Cre- Jnk1LoxP/LoxP Jnk2LoxP/LoxP Jnk3-/- (EfCtrl), and Cdh5-Cre+ Jnk1LoxP/LoxP Jnk2LoxP/LoxP Jnk3-/- (E3KO). Three separate samples from mouse lung endothelial cells of each genotype were analyzed.

Genome-wide transcriptional alterations during vascular growth in skeletal muscles of type 2 diabetic mice in response to femoral artery ligation

Project description:Diabetes is a major risk factor of cardiovascular disease including coronary and peripheral arterial disease, attributed to impaired arteriogenesis and angiogenesis. Mechanisms behind them are poorly understood due to the complexity of these processes in presence of diabetes. To understand the cellular and molecular mechanisms underlying impaired adaptive vascular responses in type 2 diabetic (T2DM) mouse models of hindlimb ischemia using Genome-wide mRNA sequencing.In response to FAL, collateral density and lumen area were significantly reduced in adductor muscles of T2DM mice at day 7 and 14 days post-FAL. mRNA sequencing suggested an altered gene expression typical for monocyte and macrophage (Mϕ) subsets. Similarly, gene expressions typical of dendritic cell and lymphocyte were altered in adductor muscles of T2DM mice. Ischemic muscles of T2DM mice displayed impaired angiogenesis as revealed by endothelial staining. mRNA sequencing suggested downregulation of crucial genes implicated in angiogenesis and differential expression of genes typical for Mϕ subsets. Immunohistochemistry analysis corroborated with the mRNA sequencing analysis, suggesting an altered Mϕ polarization behind the impaired arteriogenesis and angiogenesis seen in T2DM mouse models of hindlimb ischemia.

Improving blood vessel function in peripheral artery disease: endothelial-derived TRAIL generates stable microvessels in ischemia via TRAIL-R2

Project description:Ischemia reduction in peripheral artery disease (PAD) through angiogenesis requires endothelial cell (EC) and pericyte interactions to form stable microvascular networks, but processes mediating this are disrupted in PAD. Patients with cardiovascular disease have suppressed levels of tumour necrosis factor-related apoptosis-inducing ligand (TRAIL). The contribution of endothelial cell (EC)-derived TRAIL to generating stable blood vessel networks and the impact of its receptors in PAD is unknown.We used scRNA-seq to identify molecular determinants that are altered in ischemic tissues of EC-specifc TRAIL KO mice vs littremate control mice, specifically looking at EC-pericyte interactions.

Whole Blood Transcriptomics Differentiates Circulating Gene Expression Between Coronary Artery Disease and Peripheral Artery Disease

Project description:Coronary artery disease (CAD) and peripheral artery disease (PAD) are prevalent atherosclerotic disorders that exhibit distinct clinical and pathological presentations. We used whole-blood RNA sequencing to investigate circulating transcriptomic differences between PAD and CAD. Whole-blood RNA sequencing was performed in 71 subjects 40-65 years of age with symptomatic PAD (n=20) or CAD (n=51). Patients with concomitant PAD and CAD were excluded. Differential expression analysis was performed to compare circulating gene expression in patients with PAD and patients with CAD. We identified 106 genes differentially expressed between PAD and CAD (p adj. < 0.1). Pathway analysis identified upregulation of interferon signaling pathways in CAD and upregulation of neutrophil degranulation and toll-like receptor signaling pathways in PAD. Cell type deconvolution revealed greater fractional abundance of T cell subsets in CAD compared to PAD. Patients with PAD had higher neutrophil-lymphocyte ratio that patients with CAD (3.4 vs. 2.3, p=0.01). In conclusion, PAD and CAD have unique circulating transcriptomes characterized by differences in inflammatory gene expression.
