Effect of thermoneutral housing (as compared to room temparture) on liver gene expression in mice fed a control diet or a western diet

ABSTRACT: Liver gene expression was assessed in male mice after they had previously been fed a control diet or a western diet for 13 weeks. Mice fed the control diet were healthy. Mice fed the western diet became obese. In each diet, half of the mice were housed at room temperature (22°C) and the other half was housed at thermoneutrality (30°C). Overall, this experiment allowed us to investigate the effect of housing temperature on liver gene expression in healthy mice and in obese mice.

Next Generation Sequencing Facilitates Quantitative Analysis of epididymal adipose tissue transcriptomes from mice fed either a control-diet or aWestern-diet for one or five successive generations

Project description:Purpose: Successive paternal generations of unhealthy diet induce a fat mass increase. The goal of this study is to compare epididymal adipose tissue transcriptome profiling (RNA-seq) of mice fed either a control-Diet or a High-Fat-Diet for One or Five successive generations. The aim of the study is to identify differentially expressed RNA in epididymal adipose tissue of obese males which might be involved in the exacerbation of induced by the maintenance of High Fat Diet feeding for 5 successive generations. Methods: Epididymal adipose tissue mRNA profiles of 18-months-old mice fed either a control-Diet or a Western-Diet for One or Five successive generations were generated by deep sequencing, in triplicate, using Illumina GAIIx. The sequence reads that passed quality filters were quantitavely analyzed. Reads abundance was evaluated for each gene followed by annotation versus mouse GTF by using the featureCounts function.The R package Edger was used in order to normalize the reads and to identify differentially expressed (DE) genes. Results: RNA-seq data revealed a specific enrichment in gene related to metabolic syndrome traits in male mice obtained after successive paternal generations of Western-Diet.

Renal Proximal Tubule Cell-specific Megalin Deletion Induces Tubulointerstitial Nephritis in Mice Fed Western Diet

Project description:Pharmacological inhibition of megalin (also known as low-density lipoprotein receptor-related protein 2: LRP2) attenuates atherosclerosis in hypercholesterolemic mice. Since megalin is abundant in renal proximal tubule cells (PTCs), PTC-LRP2 +/+ and -/- littermates in an LDL receptor -/- background were generated and fed a Western diet to determine effects of PTC-derived megalin on atherosclerosis. PTC-specific megalin deletion did not attenuate atherosclerosis in LDL receptor -/- mice in either sex. Serendipitously, we discovered that PTC-specific megalin deletion led to interstitial infiltration of CD68+ cells and tubular atrophy. The pathology was only evident in male PTC-LRP2 -/- mice fed the Western diet, but not in mice fed a normal laboratory diet. Renal pathologies were also observed in male PTC-LRP2 -/- mice in an LDL receptor +/+ background fed the same Western diet, demonstrating that the renal pathologies were dependent on diet and not hypercholesterolemia. By contrast, female PTC-LRP2 -/- mice had no apparent renal pathologies. In vivo multiphoton microscopy demonstrated that PTC-specific megalin deletion dramatically diminished albumin accumulation in PTCs within 10 days of Western diet feeding. RNA sequencing analyses demonstrated the upregulation of inflammation-related pathways in kidney. Overall, PTC-specific megalin deletion leads to tubulointerstitial nephritis in mice fed Western diet, with severe pathologies in male mice.

Quercetin suppresses immune cell accumulation and improves mitochondrial gene expression in epididymal adipose tissue of diet-induced obese mice

Project description:We examined the effect of quercetin on the gene expression and function of epididymal adipose tissue (EAT) in Western diet-induced obese mice. Quercetin suppressed the increase in the number of macrophages and the decrease in the ratio of CD4+ to CD8+ T cells in EAT, and the elevation of plasma leptin and TNFÎ± levels in mice fed the Western diet. Comprehensive gene expression analysis revealed that quercetin suppressed gene expression associated with the accumulation and activation of immune cells, including macrophages and lymphocytes in EAT. It also improved the expression of the oxidative stress-sensitive transcription factor NFÎºB, NADPH oxidases, and antioxidant enzymes. Quercetin markedly increased gene expression associated with mitochondrial oxidative phosphorylation and mitochondrial DNA Quercetin most likely universally suppresses the accumulation and activation of immune cells, including anti-inflammatory cells, whereas it specifically increased gene expression associated with mitochondrial oxidative phosphorylation. Suppression of oxidative stress and NFÎºB activity likely contributed to the prevention of the accumulation and activation of immune cells and resulting chronic inflammation. Quercetin most likely universally suppresses the accumulation and activation of immune cells, including anti-inflammatory cells, whereas it specifically increased gene expression associated with mitochondrial oxidative phosphorylation. Suppression of oxidative stress and NFÎºB activity likely contributed to the prevention of the accumulation and activation of immune cells and resulting chronic inflammation. C57BL/6J mice were fed a control diet; a Western diet high in fat, cholesterol, and sucrose; or the same Western diet containing 0.05% quercetin for 18 weeks.

Quantitative proteomic analysis of oocytes from high-fat diet (HFD)-fed and normal diet mice

Project description:In order to establish an obese mouse model, female mice were continuously fed with a high-fat diet (HFD) or a normal diet (control) for 16 weeks beginning at three weeks of age. In this paper, these mice are termed ‘HFD mice’ and ‘control mice’, respectively. Accordingly, we call their oocytes ‘HFD oocytes’ and ‘control oocytes’. Substantial evidence indicates that the effects of maternal obesity on embryo/offspring development can be attributed to factors within the oocyte (9). To identify such potential effectors, we performed a comparative proteomic analysis of ovulated MII oocytes from control and HFD mice.
