Effect of NUDT21 knockdown on gene expression in MUTZ-1 and SKM-1 myelodysplastic syndrome cells

ABSTRACT: During mRNA maturation, splicing, capping and polyadenylation are its three main steps. And polyadenylation is the last key step in mRNA maturation, where the 3' end of pre-mRNA is cleaved and polyA sequence is added, which plays an important role in the stability of mRNA and its cellular function. NUDT21, which we report here, is a key regulator involved in polyadenylation in mRNA maturation and plays an extremely important role in variable polyadenylation. And we found that NUDT21 was highly expressed in MDS cells and that NUDT21 had a significant effect on the biological functions of MDS cells, especially in the transformation of MDS into AML. Therefore, our data report a specific gene profile of MDS cell lines after NUDT21 knockdown, which provides a new biomarker for clinical judgement of whether the disease has progressed in MDS patients, and provides new ideas on whether NUDT21 downstream regulatory genes, such as XAF1, may be new targets for MDS therapy.
