P5CS regulates translation by functioning as non-canonical RBP [RIP-seq]

ABSTRACT: P5CS is the rate-limiting enzyme in the proline biosynthetic pathway, while whether P5CS mediates function beyond enzyme activity remains completely unknown. Here, we reveal that P5CS functions as a non-canonical RNA-binding protein and inhibits the growth and metastasis of cancer cells by suppressing translation initiation in an enzyme activity-independent manner. To explore the underlying mechanism of P5CS in translation regulation, Ribo-seq, RIP-seq and LACE-seq were performed to investigate the downstream targets of P5CS.
