SnRNAseq from sciatic nerves from mouse of 2-3, 15-16, and 20-30 months old

ABSTRACT: Changes in gene expression during peripheral nerves aging, to the single cell level, are unkown. To explore possible age-related changes during healthy aging of peripheral nerves we compared snRNAseq of sciatic nerves from young adult (2-3 months old), senior adult (15-16 months old), and aged (20-30 month old mice.

Myelin proteome differences in old and young mice

Project description:Aging of the peripheral nervous system (PNS) is associated with structural and functional changes that lead to a reduction in regenerative capacity and the development of age-related peripheral neuropathy. Myelin is a central component in maintaining physiological peripheral nerve function, and differences in myelin maintenance, degradation, formation and clearance have been suggested to contribute to age-related PNS changes. In addition, recent proteomic studies have elucidated the complex composition of the total myelin proteome in health and its changes in neuropathy models. However, changes in the myelin proteome of peripheral nerves during ageing have not been investigated. Hence, the aim of this proteomics experiment was to define proteome changes in isolated myelin fractions during ageing, by investigating myelin proteome profiles from young (nerves from 2-3 month old mice) and old (nerves from 18 months old mice) nerves.

Transcription profiling of 8 months and 24 months old rat radial and sciatic nerves

Project description:The objective of this study was to determine which pathways are significantly regulated with age in sciatic and radial nerves, individually or considering the interaction term. We find a strong signature of the cholesterol biosynthesis pathway being downregulated with age in both nerves, however, this effect is significantly milder in the radial nerve. We collected both radial and sciatic nerves from 8 adult (8 months old) and 8 old (24 months old) rats.

Loss of adrenergic nerves in bone marrow drives hematopoietic stem cell niche aging

Project description:Aging of hematopoietic stem cells (HSCs) is associated with the decline of their regenerative capacity, and multi-lineage differentiation potential, contributing to development of blood disorders. The bone marrow microenvironment was recently suggested to influence HSC aging, however the underlying mechanisms remain largely unknown. Here, we show that HSC aging critically depends on bone marrow innervation by the sympathetic nervous system (SNS), as premature loss of SNS nerves or adrenoreceptor b3 (ADRb3) signaling in the microenvironment accelerated the appearance of HSC aging phenotypes reminiscent of physiological aging. Strikingly, supplementation of ADRb3 sympathomimetics to old mice significantly rejuvenated in vivo function of old HSCs, suggesting that the preservation or restitution of SNS innervation during aging may hold the potential for novel HSC rejuvenation strategies.

RNA sequencing of sciatic nerves from ZH3 and WT mice

Project description:The transcriptome of sciatic nerves was investigated by RNA sequencing. We compared the transcriptome of ZH3 sciatic nerves to WT sciatic nerves in two age groups. Differentially expressed genes were considered as indicators of peripheral nerve damage in ZH3 mice. Then, we compared the sciatic nerve transcriptome of ZH3 mice chronically treated with a dimeric PrP-fusion protein (FT2Fc) to the two control groups treated with either buffer or mouse IgG. This comparison revealed specific changes induced by FT2Fc treatment. Contrary to our hypothesis, FT2Fc treatment did not alter the expression of the genes that were dysregulated in young and old ZH3 mice (as identified in the first part of the study).

Genome-wide expression analysis of murine sciatic nerve of young and old mice

Project description:The regenerative capacity of peripheral nerves declines during aging, contributing to the development of neuropathies, limiting organism function. Changes in Schwann cells prompt failures in instructing maintenance and regeneration of aging nerves; altered inflammatory environment leading to a defective Schwann cell response, as an underlying mechanism of impaired nerve regeneration during aging. Chronic inflammation was detected in intact uninjured old nerves, characterized by increased macrophage infiltration and raised levels of monocyte chemoattractant protein 1 (MCP1) and CC chemokine ligand 11 (CCL11). Schwann cells in the old nerves appeared partially dedifferentiated, accompanied by an activated repair program independent of injury. Upon sciatic nerve injury, an initial delayed immune response was followed by a persistent hyperinflammatory state accompanied by a diminished repair process. As a contributing factor to nerve aging, we showed that CCL11 interfered with Schwann cell differentiation in vitro and in vivo. Our results indicate that increased infiltration of macrophages and inflammatory signals diminish regenerative capacity of aging nerves by altering Schwann cell behavior. The study identifies CCL11 as a promising target for anti‐inflammatory therapies aiming to improve nerve regeneration in old age.

Multi-omic characterization of human sural nerves across polyneuropathies [Xenium]

Project description:Diseases of peripheral nerves termed polyneuropathies (PNPs) are common, mechanistically heterogeneous, and challenging to diagnose. Here, we integrated single nucleusi transcriptomics of peripheral nerves from 33 human PNP patients and four controls (365,708 nuclei) with subcellular spatial transcriptomics. We identified novel and human-specific nerve cell type markers including unexpectedly heterogeneous perineurial fibroblasts. All PNPs shared a loss of myelinating and an increase in repair Schwann cells and endoneurial lipid-associated macrophages. Transcriptional changes affected multiple cells outside of the endoneurium across PNPs, suggesting PNPs as ‘pan-nerve diseases’. Spatially, PNPs showed a previously unknown perineurial hyperplasia and fibrotic dispersion and this was most pronounced in immune-mediated PNPs. Single cell transcriptomics supported the differential diagnosis of PNPs with potential for future unbiased diagnostic classification.
