BiTE-secreting T cells rationally combine with PD-1 blockade and promote remodeling of the local immune landscape to enhance response durability in ovarian cancer

ABSTRACT: scRNA-seq (5' GEX) was used to examine the peritoneal TME in IE9-mp1-hFRa cells bearing animals treated with FRB-T cell + anti-PD1 therapies. We observed unique TME composition associated with acute and durable responses to combination therapy that was disrupted in progressive disease. Further we found that providing an AD-OVA immunization boost along the course of therapy enhanced favorable TME composition and overall tumor control.

BiTE-secreting T cells effectively target ovarian cancer and prolong antitumor immunity by persisting in the extratumoral peritoneal cavity

Project description:Cancer immunotherapies can produce complete therapeutic responses, however outcomes in ovarian cancer (OC) are modest and while adoptive transfer of engineered T cells (ACT) has been evaluated in OC, durable effects are rarely observed. T cells lacking tumor specificity (bystander T cells) readily accumulate in tumors and leveraging bystander T cell activity represents a promising opportunity to enhance antitumor immunity following ACT. To this end, we have generated T cells that stably secrete a FRα-directed bispecific T cell engager (FR-B T cells), which robustly targeted FRα+ tumor cells in OC patient specimens and effectively engaged patient T cells present in the tumor microenvironment (TME). Therapeutic testing of FR-B T cells using an immunocompetent OC model demonstrated robust activity, with response duration dependent on both endogenous T cells and FR-B T cell persistence. Cytokine preconditioning prior to infusion produced less differentiated FR-B T cells and improved therapeutic efficacy, with mechanistic studies revealing preferential accumulation of TCF-1+ CD69-CD39- stem-like CD8+ FR-B T cells in the peritoneal cavity over solid tumors. These findings highlight the potential of FR-B T cell therapy in OC and suggest the peritoneal TME permits FR-B T cells to persist in the extratumoral space while actively directing antitumor immunity.

Triple Stiffness: A Bioinspired Strategy to Combine Load-Bearing, Durability, and Impact-Resistance.

Project description:Structures with variable stiffness have received increasing attention in the fields of robotics, aerospace, structural, and biomedical engineering. This is because they not only adapt to applied loads, but can also combine mutually exclusive properties. Here inspired by insect wings, the concept of "triple stiffness" is introduced and applied to engineering systems that exhibit three distinct deformability regimes. By implementing "flexible joints," "mechanical stoppers," and "buckling zones," structures are engineered to be not only load-bearing and durable, but also impact-resistant. To practice the performance of the design concept in real-life applications, the developed structures are integrated into 3D printed airplane wing models that withstood collisions without failure. The concept developed here opens new avenues for the development of structural elements that are load-bearing, durable, and impact-resistant at the same time.

Bone marrow-derived multipotent mesenchymal stromal cells promote tongue cancer invasion by inducing collagen production

Project description:Tumor microenvironment (TME) is an active player in malignant growth and spread. Changes in the composition and structure of TME and extracellular matrix can result in either suppression or facilitation of malignant tumor growth. Carcinoma‐associated fibroblasts, bone marrow-derived multipotent mesenchymal stromal cells (BMMSCs), tumor associated macrophages and other inflammatory cells all affect the composition of TME, proliferation and survival of cancer cells, angiogenesis, invasion and metastasis. The objective of this work was to investigate the effect of the interaction between bone marrow-derived BMMSCs and human oral tongue squamous cell carcinoma (OTSCC) cells in the processes of invasion and gene expression. Co-cultures of OTSCC cancer cells and BMMSCs in 3D organotypic invasion assay were used in addition to cell culture, immunological, microarray, and RNA interference techniques. Total number of 4 samples were analyzed. 2 replicates of cultured human oral tongue squamous cell carcinoma (OTSCC) cells, and 2 replicates of OTSCC cells co-cultured with bone marrow-derived multipotent mesenchymal stromal cells

A genome wide RNAi screen in mouse embryonic stem cells identifies Mp1 as a key mediator of differentiation

Project description:Despite intense investigation of intrinsic and extrinsic factors that regulate pluripotency, the process of initial fate commitment of embryonic stem (ES) cells is still poorly understood. Here, we used a genome wide shRNA screen in mouse ES cells to identify genes that are essential for initiation of differentiation. Knockdown of the scaffolding protein Mek binding protein 1 (Mp1, also known as Lamtor3, Map2k1ip1) stimulated self-renewal of ES cells, blocked differentiation and promoted proliferation. Fibroblast growth factor 4 (FGF4) signaling is required for initial fate commitment of ES cells. Knockdown of Mp1 inhibited FGF4-induced differentiation but did not alter FGF4 driven proliferation. This uncoupling of differentiation and proliferation was also observed when oncogenic Ras isoforms were over expressed in ES cells. Knockdown of Mp1 redirected FGF4 signaling from differentiation towards pluripotency and upregulated the pluripotency-related genes Esrrb, Rex1, Tcl1 and Sox2. We also found that human germ cell tumors express low amounts of Mp1 in the invasive embryonic carcinoma and seminoma histologies and higher amounts of Mp1 in the non-invasive carcinoma in situ precursor and differentiated components. Knockdown of Mp1 in invasive germ cell tumor cells resulted in resistance to differentiation, thereby showing a functional role for Mp1 both in normal differentiation of ES cells as well as in germ cell cancer.

BiTE-Secreting CAR-γδT as a Dual Targeting Strategy for the Treatment of Solid Tumors.

Project description:HLA-G is considered as an immune checkpoint protein and a tumor-associated antigen. In the previous work, it is reported that CAR-NK targeting of HLA-G can be used to treat certain solid tumors. However, the frequent co-expression of PD-L1 and HLA-G) and up-regulation of PD-L1 after adoptive immunotherapy may decrease the effectiveness of HLA-G-CAR. Therefore, simultaneous targeting of HLA-G and PD-L1 by multi-specific CAR could represent an appropriate solution. Furthermore, gamma-delta T (γδT) cells exhibit MHC-independent cytotoxicity against tumor cells and possess allogeneic potential. The utilization of nanobodies offers flexibility for CAR engineering and the ability to recognize novel epitopes. In this study, Vδ2 γδT cells are used as effector cells and electroporated with an mRNA-driven, nanobody-based HLA-G-CAR with a secreted PD-L1/CD3ε Bispecific T-cell engager (BiTE) construct (Nb-CAR.BiTE). Both in vivo and in vitro experiments reveal that the Nb-CAR.BiTE-γδT cells could effectively eliminate PD-L1 and/or HLA-G-positive solid tumors. The secreted PD-L1/CD3ε Nb-BiTE can not only redirect Nb-CAR-γδT but also recruit un-transduced bystander T cells against tumor cells expressing PD-L1, thereby enhancing the activity of Nb-CAR-γδT therapy. Furthermore, evidence is provided that Nb-CAR.BiTE redirectes γδT into tumor-implanted tissues and that the secreted Nb-BiTE is restricted to the tumor site without apparent toxicity.

Project description:With the advent of cancer immunotherapy, intense investigation has been focused on tumor-infiltrating immune cells. With only a fraction of patients responding to these new therapies, a better understanding of all elements of the tumor microenvironment (TME) that may influence therapeutic outcome is needed. Stromal elements of the TME, chiefly fibroblasts, have emerged as potential contributors to tumor progression and most recently resistance to immunotherapy, but their precise composition and clinical relevance remain incompletely understood. Here we use single-cell transcriptomics to chart the fibroblastic landscape during pancreatic ductal adenocarcinoma (PDAC) progression in animal models, identifying two healthy tissue fibroblast subsets that co-evolve along individual trajectories into four subsets of carcinoma-associated fibroblasts (CAFs).
