Single-cell transcriptomics reveals markers of regulatory T cell dysfunction in Gestational Diabetes Mellitus

ABSTRACT: Gestational Diabetes Mellitus (GDM) is a common pregnancy complication and the role of regulatory T cells (Tregs) in this disease is ambiguous. This study aims to determine the aberrant transcriptional landscape in Tregs from GDM patients and controls. We identify gene expression programmes dysregulated in GDM Tregs and assess their utlilty as GDM biomarkers in bulk mRNA.

Placenta-derived exosomes of gestational diabetes mellitus affect cell metabolism and cell function via transferring microRNAs

Project description:Gestational Diabetes Mellitus (GDM) is a common pregnancy complication and exosomal-miRNAs played a critical role in the development of GDM. Our research aimed to investigate the mechanism of exosomes participation in GDM and proposed the innovative therapeutic methods. In the present study, we isolated and identified the placenta-derived exosomes from GDM patients. Exosomes could be internalized by HTR8 cells and induced the lipid metabolism dysfunction. Differential metabolites in placenta tissues of GDM patients were analyzed and lipid metabolites occupied the highest proportion. Besides, exosomes promoted the oxidative stress production and inhibited cell autophagy and promoted cell apoptosis by transferring miR-152-5p. In addition, miR-152-5p inhibitor could reverse the ROS levels and regulate cell autophagy and cell apoptosis. Therefore, miR-152-5p inhibitor might be an efficient therapy for GDM. Our findings revealed that placenta-derived exosomes of GDM could regulate affect cell lipid metabolism, increase oxidative stress and regulate cell autophagy and apoptosis. Exosomal miR-152-5p is expected to be an promising target for GDM.

Gut Microbiome and Serum Metabolic Alterations in Recurrent Gestational Diabetes Mellitus

Project description:<p>Gestational diabetes mellitus (GDM) is a common complication of pregnancy, particularly affecting multiparous women with a history of GDM, who face a recurrence risk approaching 50%. This study investigates the association between gut microbiota modifications and metabolic profiles from early to mid-pregnancy and the recurrence of GDM in a cohort of 70 multiparous women with prior GDM. Conducted as a prospective cohort study at Beijing Obstetrics and Gynecology Hospital from July 2018 to December 2020, participants underwent assessments including oral glucose tolerance tests (OGTT) and collection of fecal and serum samples. Microbial DNA was extracted, with the V3-V4 region of the 16S rRNA gene sequenced, while serum metabolites were analyzed through non-targeted metabolomics using mass spectrometry. Results indicated significant alterations in gut microbiota composition, with specific genera associated with increased GDM risk. Notably, the study identified distinct metabolic patterns, revealing downregulation of several key metabolites in recurrent GDM cases compared to controls. Pathway enrichment analysis highlighted dysregulation in metabolic functions pertinent to serotonergic synapses, arachidonic acid metabolism, unsaturated fatty acid biosynthesis, and caffeine metabolism. Overall, this research underscores the critical role of gut microbiota and metabolic changes in understanding GDM recurrence, providing insights that may inform targeted preventive strategies.</p>

Panoramic Elucidation of the Molecular Mechanisms Underpinning the Epigenetic Impact of Gestational Diabetes Mellitus on Offspring

Project description:Background: Gestational diabetes mellitus (GDM), a common pregnancy complication with profound effects on maternal and offspring health, involves epigenetic modifications, particularly DNA methylation, but the molecular mechanisms elevating metabolic disease risk in offspring remain elusive. Methods: Integrating public data with whole-genome methylation data from umbilical cord blood of GDM fetuses as the validation cohort, this study characterizes GDM-associated epigenetic features and develops a predictive model validated across external datasets to assess potential long-term health risks in offspring. Results:We identified consistent patterns of differential methylation in the offspring of GDM, with key DMPs associated with glucose homeostasis and insulin sensitivity genes. Pathway analysis revealed enrichment in insulin signaling, AMPK activation, and adipocytokine signaling pathways. Our predictive model demonstrated robust performance across different sample sizes (AUC=0.89 in public data, AUC=0.82 in validation cohort). CpG sites associated with PPARG and INS genes showed persistent predictive significance across datasets. Conclusions:This study provides a comprehensive characterization of the epigenetic landscape in the offspring of GDM, revealing consistent methylation patterns and pathway enrichments across diverse datasets. The developed predictive model holds potential for assessing health risks of GDM in offspring, advancing understanding of GDM pathophysiology and offering epigenetic biomarkers for future diagnostic, therapeutic, and preventive strategies.

Plasma exosomal microRNAs associated with metabolism as early predictor of gestational diabetes mellitus

Project description:The microRNA (miRNA) expression profile of plasma exosome in pregnant women complicated with gestational diabetes mellitus (GDM) has not been fully clarified. In this study, differentially expressed miRNAs in plasma exosomes were identified by high-throughput small RNA sequencing in 12 GDM and 12 normal glucose tolerance (NGT) pregnant women and validated in 102 GDM and 101 NGT pregnant women. A total of 22 exosomal miRNAs were found and five of them were verified by qRT-PCR. Exosomal miR-423-5p was upregulated, while miR-99a-5p, miR-192-5p, miR-148a-3p, and miR-122-5p were downregulated in pregnant women complicated with GDM.
