Effects of interference and overexpression of NUDT3 on fat deposition in porcine subcutaneous adipocytes of pigs

ABSTRACT: The enforced expression of NUDT3 significantly suppressed adipogenesis and lipid accumulation in porcine subcutaneous adipocytes. In addition, exogenous expression of NUDT3 in adipose tissue significantly reduced fat expansion triggered by a high-fat diet in mice. Mechanistically, the DEGs affected by NUDT3 overexpression and interference were significantly enriched in the PI3K-Akt signaling pathway were revealed. Western blotting further confirmed that the phosphorylation of Akt was significantly increased by NUDT3 knockdown, and the levels of phosphorylated PI3K, Akt and mTOR were significantly decreased by the enforced expression of NUDT3 both ex vivo and in vivo.
