ABSTRACT: Transgelin expression in colorectal cancer has been reported to correlate with patient prognosis. The aim of this study was to investigate the function of Transgelin in fibroblasts of colorectal cancer. As a method, we assessed whether knockdown of Transgelin alters the expression of the gene.

Quantitative proteomics reveals that distant recurrence-associated protein R-Ras and Transgelin predict post-surgical survival of colorectal cancer Stage III patients

Project description:Surgical resection is the major clinical intervention for Stage III colorectal cancer (CRC) currently. However, as much as 30.8% of the patients who had ever taken curative resection came out of recurrence eventually. Therefore, to facilitate formulating effective treatment plans, there is an intense demand for Stage III CRC post-surgical prognostic biomarkers. In this study, we identified total 146 differentially expressed proteins (DEPs) associated with poor prognosis in Stage III CRC patients with TMT-based quantitative mass spectrometry (MS). In these DEPs, the protein expression level of R-Ras and Transgelin were tested with immunohistochemistry (IHC) of 192 individual specimens. Further Kaplan-Meier analysis revealed that the level of R-Ras and Transgelin is associated with patients’ 5-year overall survival (OS) and disease-free survival (DFS) significantly, and multivariate Cox-regression analyses revealed that R-Ras and Transgelin are independent prognostic factors for OS and DFS respectively. In conclusion, our study presents that R-Ras and Transgelin are potential post-surgical prognostic biomarkers of Stage III CRC.

Expression data of TAGLN-overexpressing RKO human colon cancer cells

Project description:Transgelin was the top-ranked marker of metastatic potential identified in the comparison of node-positive colorectal cancer (CRC) versus node-negative CRC in our previous study. Transgelin is localized in the nucleus of cultured CRC cells and microRNA-mediated knockdown of TAGLN (the gene encoding transgelin) expression modulates the expression of genes involved in the epithelial-to-mesenchymal transition. We performed gene expression profiling on control and transgelin-overexpressing RKO cells using Affymetrix microarray technology. The plasmid carrying TAGLN was generated using the pcDNA6.2/EmGFP-Bsd/V5-DEST vector (Invitrogen, Carlsbad, CA) and the pDONR221-TAGLN-mut plasmid from a previous study by an LR recombination reaction (http://www.invitrogen.com). After lipofectamine-mediated transfection of the TAGLN-carrying plasmid or the vector-only control plasmid, stable transfectants were selected and cultured in medium containing 25 Î¼g/ml blasticidin followed by RNA extraction and hybridization on Affymetrix microarrays. Six samples were obtained from three independent experiments (technical replicates).

Project description:The clinical management of colorectal cancer patients has significantly improved due to the identification of new chemotherapeutic targets. Since rapid tumor proliferation is associated with poor patient prognosis, here we characterize the transcriptional signature of rapidly proliferating colorectal cancer cells in an attempt to identify novel candidate therapeutic targets. Therefore, the doubling time of 52 colorectal cancer cell lines was determined and genome-wide expression profiling of a subset of 31 of these cell lines was assessed by microarray analysis.

Transgelin silencing induces different processes in differrent breast cancer cell lines

Project description:Transgelin is a protein which has been described as a marker of several cancers. Interestingly, the literature describes both up- and down-regulation of transgelin in the tumor in comparison with non-tumor tissue. The mechanisms of transgelin function in cancer are considerably unknown. Transgelin is abundant especially in smooth muscle cells. It is associated with actin stress fibers. These contractile structures participate among others in cell motility, adhesion or morphology. We focused on transgelin function in breast cancer. Initially, we studied transgelin role in cell migration in the breast cancer cell lines BT 549 and PMC 42. Using xCELLigence system we reached opposite results in two cell lines. Transgelin silencing increased and decreased migration of PMC 42 and BT 549 cells, respectively. To further clarify these contradictory results, we performed an experiment to quantify proteomic changes after transgelin silencing in these two cell lines using quantitative proteomics (iTRAQ-2DLC-MS/MS). Our results confirmed transgelin function in the migration of BT 549 cells and suggested transgelin role in apoptosis and small molecule biochemistry in PMC 42 cells.
