Project description:N. oceanica IMET1 was inoculated into modified f/2 liquid medium, which was prepared with 35 g L−1 sea salt, 1 g L−1 NaNO3, 67 mg L−1 NaH2PO4*H2O, 3.65 mg L−1 FeCl3*6H2O, 4.37 mg L−1 Na2EDTA*2H2O, trace metal mix (0.0196 mg L−1 CuSO4*5H2O, 0.0126 mg L−1 NaMoO4*2H2O, 0.044 mg L−1 ZnSO4*7H2O, 0.01 mg L−1 CoCl2, and 0.36 mg L−1 MnCl2*4H2O), and vitamin mix (2.5 µg L−1 VB12, 2.5 µg L−1 biotin, and 0.5 µg L−1 thiamine HCl). The algal cells were grown in liquid cultures under continuous light (∼500 µmol photons m−2 s−1) at 25°C and aerated by bubbling with a mixture of 1.5% CO2 in air.

Project description:Nannochloropsis oceanica CCMP1779 is a marine unicellular stramenopile and an emerging reference species for basic research on oleogenic microalgae with biotechnological relevance. We investigated its physiology and transcriptome under light/dark cycles. We observed oscillations in lipid content and a predominance of cell division in the first half of the dark phase. Globally, more than 60% of the genes cycled in N. oceanica CCMP1779, with gene expression peaking at different times of the day. Interestingly, the phase of expression of genes involved in certain biological processes was conserved across photosynthetic lineages. Furthermore, in agreement with our physiological studies we found the processes of lipid metabolism and cell division enriched in cycling genes. For example, there was tight coordination of genes involved in the lower part of glycolysis, fatty acid synthesis and lipid production at dawn preceding lipid accumulation during the day. Our results suggest that diel lipid storage plays a key role for N. oceanica CCMP1779 growth under natural conditions making this alga a promising model to gain a basic mechanistic understanding of triacylglycerol production in photosynthetic cells. Our data will help the formulation of new hypotheses on the regulation transcriptional control of cell growth and metabolism in Nannochloropsis. Nannochloropsis oceanica CCMP was entrained to 12:12 light:dark cycles and biological replicates collected every 3 hours for a cycle for a total of 16 samples.

Project description:Light quality is an important abiotic factor that affects growth and development of photosynthetic organism. In this study, D. salina was exposed to red (660 nm) and blue light (450 nm), and cell growth, pigments, and transcriptome were analyzed. The RNA of D. salina was sequenced and transcriptomic response of algal cells after transitioning from white light to red and blue light was investigated. Genes encoding for enzymes involved in photosynthesis were down-regulated, whereas genes involved in the metabolism of carotenoid were up-regulated. Genes encoding for photoprotective enzymes related to reactive oxygen species scavenging were up-regulated under both red and blue light. The present transcriptomic study would assist in the comprehensive understanding of carotenoid biosynthesis of D. salina.

Phytochrome-Interacting bHLH transcription Factors (PIFs), which are repressors of photomorphogenesis accumulate in darkness and are degraded in the light in a phytochrome-dependent manner. Two PIFs, PIF1 and PIF3 accumulated in the light in hmr mutants. In order to determine the gene expression of PIF-dependent genes in hmr mutants in the light, we have performed whole-genome expression analysis on two hmr mutants: a null allele, hmr-5; and a weak allele, hmr-22.

Project description:We investigated light dependent gene expression changes in the marine ochrophyte Nannochloropsis oceanica CCMP1779. These algae have several putative blue light photoreceptors but appear to lack red light photoreceptors. To study early light signaling in N. oceanica and avoid as much as possible secondary downstream events, we quantified gene expression changes in dark-adapted cells after a short blue or red light pulse. More genes were differentially expressed under blue than under red light. In addition, fold change in expression was smaller for the red light-treated samples. For example, the median fold change of induced genes was 3 for blue light and 2.5 for red light. Moreover, hierarchical cluster analysis showed that gene expression after red light treatment was more similar to the dark control than after blue light treatment.

Project description:Myxobacteria are social bacteria distinguished among prokaryotes by a multicellular stage in their life cycle known as fruiting bodies formations, which is a response to nutrient deprivation. Additionally, fruiting body formation in Stigmatella aurantiaca is stimulated by light. Genome sequence of Myxococcus macrosporus, revealed the presence of a red-light photoreceptor, bacteriophytochrome (BphP), found in photosynthetic and non-photosynthetic bacteria. BphPs belong to a large phytochrome family of photoreceptors originally discovered in plants, where they are directly linked to the circadian clock genes. BphPs have been implicated in carotenoid synthesis, motility, and photodynamic stress in non-photosynthetic bacteria. The role of the BphPs in M. macrosporus remains unknown although it has been linked to fruiting body formation in related S. aurantiaca. We have observed that M. macrosporus displays differential photomorphogenic behaviors under constant light, and 12:12 hour light/dark (LD) cycle as compared to constant dark (DD) conditions. We hypothesize that BphP plays a role in these behaviors. In this study we demonstrate light and nutrient dependent pattern of fruiting bodies development in Myxococcus macrosporus HW-1. Additionally, we characterized light induced differential gene expression with the special focus on the circadian clock, and carotenoid biosynthesis genes in M. macrosporus and M. xanthus.
