Single cell analysis reveals the myeloid-biased hematopoiesis and leukemic evolution programs initiated by Brd4 in HSPCs of mice with disuse osteoporosis

ABSTRACT: To understand the molecular changes driving the myeloid-biased hematopoiesis and the potential of leukemia evolution by restricting the differentiation and apoptosis in HSPCs of OP-HU mice, single cell RNA sequencing was performed to analyze the transcriptional changes of Lin-/c-kit+ HSPCs from the femur and tibia of OP-HU and control mice.

Identification of cytokine signals regulated by Caspase-9

Project description:Genome-wide transcriptional profiles of the hematopoietic stem and progenitor cell (HSPCs)-enriched population derived from bone marrow chimeras reconstituted with Caspase-9 or Bak and Bax deficient fetal liver cells. Gene ontology analysis revealed changes in the expression of genes associated with interferon responses. HSPCs (Lineage-Kit+CD45.2+) were flow sorted from the femur and tibia of bone marrow chimeras 12-16 weeks post transplant of 2 million embryonic day (E) 13.5 fetal liver cells. Total RNA was isolated and up to 250 ng of total RNA per sample was labelled and hybridized to Illumina Mouse WG6V2 expression BeadChip platform. Three independent samples were used for each population.

Expression data from growth plate cartilage of control and immobilized embryonic chickens

Project description:Affymetrix Chicken Gene 1.0 ST Array (ChiGene-1_1-st) profiles were generated from growth plate cartilage derived from the distal femur and proximal tibia of embryonic chickens at three developmental stages (day 12, 13 and 15) corresponding to stages prior to and immediately after the acqusition of mechano-responsive cartilage growth in the femur but not tibia (which does not respond to mechanical stimuli until later stages). Additionally, array profiles were generated from growth plate cartilage from the distal femur and proximal tibia of E15 chicks which were pharmacologically immobilized by decamethonium bromide treatment, which reduces longitudinal growth of the femur only at this stage.

The Link between Osteoporosis and Frozen Shoulder: Exploring the Therapeutic Effect of TAK715 on Reversing Fibrosis and Protecting against Osteoporosis via the p38 MAPK Signaling Pathway

Project description:The global incidence of frozen shoulder (FS) and osteoporosis (OP) is high. Although FS patients may experience localized OP in the shoulder, there is still insufficient strong evidence to confirm the relationship between FS and OP. The main objective of this current research is to investigate the pathogenesis mechanism of FS and explore the connection between FS and OP.We obtained FS and OP datasets from GEO and identified crosstalk genes. We screened the p38 MAPK signaling pathway and its specific inhibitor, TAK715. We conducted flow cytometry, western blot, and PCR analyses to assess the treatment effect of TAK715 on FS synovium fibroblasts at concentrations of 1μM, 5μM, and 10μM. Additionally, we employed SD rats to validate the treatment effects of TAK715 in vivo. TAK715 was found to also intervene in the activation of osteoclasts. We successfully established a FS and OP rat model, with the FS with OP rat displaying more severe symptoms. Results demonstrated that TAK715 effectively corrected both fibrosis and osteoporosis simultaneously in vivo.The frozen shoulder with osteoporosis may exhibit more severe symptoms, and TAK715 is effective in protecting fibrosis and osteoporosis both in vitro and vivo. The therapy to correct FS and OP simultaneously by TAK715 provides novel approach in FS treatment and study.

Project description:Bone marrow Hdc-GFPhi and Hdc-GFPlo HSPC (SLAM-LSK, Lin-c-kit+Sca-1+CD150+CD48-) HSCs were isolated from mouse femur and tibia from histidine decarboxylase (Hdc) green fluorescent protein (Hdc-GFP) mice. Hdc-GFPhi HSC and Hdc-GFPlo HSC cells were sorted by combinations of GFP and the cell surface markers of HSC. total RNA was isolated from sorted 2,500 HSPCs using ARCTURUS PicoPure RNA isolation kit (Life Technologies). cDNA was amplified and libraries were constructed by using SMARTer Ultra Low Input RNA kit (Clontech Laboratories) and Nextera XT DNA Library Preparation kit (Illumina) according to manufacturer’s instructions respectively. Sequencing was performed on the Illumina HiSeq2000 platform.
