Spatial transcriptomic analysis of developing hearts at embryonic and neonatal stages

ABSTRACT: Cd1 wildetype mice, Wt1-CreER;Rosa-mTmG mice, and Wt1-CreER;Rosa-DTA mice derived hearts were isolated and directly embeded. The hearts were then sectioned for spatial transcriptomic analysis.

The single cell RNA seq of pulmonary alveolar type II (AT2) cells

Project description:The pulmonary alveolar epithelium mainly composed of two types of epithelial cells: alveolar type I (AT1) and type II (AT2) cells. AT2 cells are the alveolar stem cells, and can differentiate into AT1 cells post-pneumonectomy (PNX). Here, we found that, compared with control mice (Sftpc-CreER; Cdc42flox/+; Rosa26-mTmG) at post-PNX day 21, Cdc42 AT2 null mice (Sftpc-CreER; Cdc42flox/-; Rosa26-mTmG) at post-PNX day 21 undergone fibrotic change. By using 10X genomics “Chromium Single Cell” technology, we performed single-cell RNA-seq analyses of AT2 cells of sham treated control mice (C0), AT2 cells of control mice at post PNX day 21 (C21) , AT2 cells of sham treated Cdc42 AT2 null mice (N0), and AT2 cells of Cdc42 AT2 null mice at post PNX day 21 (N21). The study identified a specific gene signature in AT2 cells of Cdc42 AT2 null mice at post PNX day 21 which is related to the fibrosis phenotype of Cdc42 AT2 null mice.

Transcription profiling of synovial sarcoma-like tumors induced in a genetically engineered mouse model

Project description:Synovial sarcoma-like tumors were generated in mice by conditionally expressing the human t(X;18) translocation-derived SYT-SSX2 fusion protein. Using a Tamoxifen-inducible CreER system, we show here that sporadic expression of SYT-SSX2 across multiple tissue types leads to exclusive formation of synovial sarcoma-like tumors while its widespread expression is lethal. CreER-based sporadic expression both avoids the severe early developmental phenotypes associated with widespread SYT-SSX2 expression and better models natural pathogenesis of cancers where transformed cells usually arise within an environment of largely normal cells. Experiment Overall Design: Genetically engineered mice capable of conditionally expressing the human synovial sarcoma-associated SYT-SSX2 fusion oncogene were mated with genetically engineered mice expressing the CreER fusion protein from ROSA locus. The progenies harboring both CreER and SYT-SSX2 were followed up with or without tamoxifen injection. Tumors were generated in these mice that were dissected out, RNA extracted, and subjected to expression profiling by microarray analysis.

Identification of the gene profiling of Wt1GFP expressing cardiomyocytes at different stages of embryonic heart development [M]

Project description:Detailed FACS analyses of enzymatically disaggregated embryonic hearts from Wt1GFPKI mice revealed two distinct types of Wt1-GFP-positive cells based on their GFP expression levels: a Wt1-GFP bright population (H) and a Wt1-GFP dim (M) population. Interestingly, when viewed under the microscope, cultured Wt1GFP+ (M) cells were shown to exhibit synchronous contractions that suggest a cardiomyocyte identity. These observations also imply that a population of low Wt1-expressing cardiomyocytes contributes to the formation of the myocardium.

Gene expression profiling of Scgb1a1+, CD24low cells isolated from the lungs of Scgb1a1-CreER; Rosa26-mTmG mouse.

Project description:We established a new method to find out subpopulation of lung epithelial stem cells, named scMORN, and uncovered the existence of Club cell subpopulations contributing to alveolar regeneration. We isolated entire Club cells by sorting Scgb1a1+, CD24low cells from the lungs of Scgb1a1-CreER; Rosa26-mTmG mouse. The gene expression profiling revealed that two genetically distinct types of Club cells.

Project description:Notch signaling defines a conserved, fundamental pathway, responsible for determination in metazoan development and is widely recognized as an essential component of lineage specific differentiation and stem cell self-renewal in many tissues including the hematopoietic system. Until recently, the majority of studies in the hematopoietic system focused on Notch signaling in lymphocyte differentiation and knowledge of individual Notch receptor roles in early hematopoiesis has been limited due to a paucity of genetic tools available To fate-map Notch receptor expression and pathway activity in the hematopoietic system we used tamoxifen-inducible CreER knock-in mice for individual Notch receptors in combination to a novel Notch reporter strain (Hes1GFP) and a conditional gain of function allele of Notch2 receptor (Rosa-lsl-ICN2). Bone marrow lineage negative, cKit+, Sca1- cells were sorted from Rosa-lsl-ICN2 Mx1-cre+ mice or Mx1-cre+ littermates for RNA extraction and hybridization on Affymetrix microarrays
