Project description:Introduction: The analysis of post-mortem metabolomic changes in biological fluids opens the way to develop new methods for the estimation of post-mortem interval (PMI). It may also help in the analysis of disease-induced metabolomic changes in human tissues when the postoperational samples are compared to the post-mortem samples from healthy donors. Objectives: The goals of this study are to observe and classify the post-mortem changes occurring in the rabbit blood, aqueous and vitreous humors (AH and VH), to identify the potential PMI markers among a wide range of metabolites, and also to determine which biological fluid – blood, AH or VH – is more suitable for the PMI estimation. Methods: The quantitative metabolomic profiling of samples of the rabbit serum, AH and VH taken at different PMIs has been performed with the combined use of high-frequency NMR and high-resolution LC-MS methods. Results: The quantitative levels of 63 metabolites in the rabbit serum, AH and VH at different PMIs have been measured. It has been found that the post-mortem metabolomic changes in AH and VH proceed slower than in blood, and the data scattering is lower. Among the metabolites whose concentrations increase with time, the most significant and linear growth is found for hypoxanthine, choline and glycerol. Conclusion: The obtained results indicate the advantage of ocular fluids, AH and VH, over blood serum for the search of potential biochemical markers for the PMI estimation. Among the compounds studied in the present work, hypoxanthine, choline and glycerol give the biggest promise as the potential PMI biomarkers.

Project description:Accurate estimation of the postmortem interval is critical in forensic investigations to determine the time since death. While traditional methods offer limited precision, molecular approaches based on gene expression have shown promise. mRNA retains detectable and quantifiable expression changes that reflect biological processes occurring after death. In this study, we analyzed the transcriptome of rat skeletal muscle at 0 hours (control) and 48 hours of post mortem interval to identify differentially expressed mRNAs that may serve as potential biomarkers, aiming to enhance the accuracy and reliability of PMI estimation in forensic science. Microarray analysis using the Affymetrix Clariom S Rat array (Affymetrix) facilitated a comprehensive overview of the skeletal muscle transcriptome at 48 hours of postmortem interval. This platform allowed for the simultaneous quantification of thousands of transcripts, revealing widespread gene expression alterations associated with the postmortem interval. The resulting transcriptomic profile not only provided insight into molecular changes occurring after death but also highlighted specific mRNAs as potential biomarker candidates for PMI estimation in forensic contexts.

Project description:The post-mortem interval (PMI) is the time that elapses since the death of an individual until the body is found. Different molecules have been analyzed to better estimate the PMI with variable results. The miRNAs draw attention in the forensic field to estimate the PMI, since they can better support degradation. To find potential biomarkers for PMI estimation, we analyzed the miRNome at early PMI in rat skeletal muscle using the Affymetrix GeneChip™ miRNA 4.0 micoarrays. In this dataset, we include the expression of 1218 rat miRNAs at early postmortem interval.

Project description:<p><strong>INTRODUCTION:</strong> Application of metabolomic methods to forensic studies may expand the limits of the post-mortem interval (PMI) estimation, and improve the accuracy of the estimation. To this end, it is important to determine which tissue is the most suitable for analysis, and which compounds are the most promising candidates for PMI estimation.</p><p><strong>OBJECTIVES:</strong> This work is aimed at the comparison of human serum, aqueous humor (AH), and vitreous humor (VH) as per- spective tissues for metabolomic-based PMI estimation, at the determination of most promising PMI biomarkers, and at the development of method of PMI estimation based on the measurement of concentrations of PMI biomarkers.</p><p><strong>METHODS:</strong> Quantitative metabolomic pro ling of samples of the human serum, AH, and VH taken at di erent PMIs has been performed with the use of NMR spectroscopy.</p><p><strong>RESULTS:</strong> It is found that the metabolomic changes in anatomically isolated ocular uids are slower and smoother than that in blood. A good positive time correlation (Pearson coe cient r > 0.5) was observed for several metabolites, including hypox- anthine, choline, creatine, betaine, glutamate, and glycine. A model for PMI estimation based on concentrations of several metabolites in AH and VH is proposed.</p><p><strong>CONCLUSIONS:</strong> The obtained results demonstrate that the metabolomic analysis of AH and VH is more suitable for the PMI estimation than that of serum. The compounds with good positive time correlation can be considered as potential PMI biomarkers.</p><p><br></p>

Project description:An estimation of the post mortem interval (PMI) is frequently touted as the Holy Grail of forensic pathology. During the first hours after death, PMI estimation is dependent on the rate of physical observable modifications including algor, rigor and livor mortis. However, these assessment methods are still largely unreliable and inaccurate. Alternatively, RNA has been put forward as a valuable tool in forensic pathology, namely to identify body fluids, estimate the age of biological stains and to study the mechanism of death. Nevertheless, the attempts to find correlation between RNA degradation and PMI have been unsuccessful. The aim of this study was to characterize the RNA degradation in different post mortem tissues in order to develop a mathematical model that can be used as coadjuvant method for a more accurate PMI determination. For this purpose, we performed an eleven-hour kinetic analysis of total extracted RNA from murine's visceral and muscle tissues. The degradation profile of total RNA and the expression levels of several reference genes were analyzed by quantitative real-time PCR. A quantitative analysis of normalized transcript levels on the former tissues allowed the identification of four quadriceps muscle genes (Actb, Gapdh, Ppia and Srp72) that were found to significantly correlate with PMI. These results allowed us to develop a mathematical model with predictive value for estimation of the PMI (confidence interval of ±51 minutes at 95%) that can become an important complementary tool for traditional methods.

Project description:This study investigates mRNA degradation in human dental pulp to explore its utility for estimating the late postmortem interval (LPMI). Morphological changes in pulp tissue at 0, 7, 14, 21 and 28 days postmortem were observed via HE staining, showing progressive cellular degradation. High-throughput sequencing of samples at 0, 7, and 21 days identified candidate mRNAs. Five mRNA biomarkers were obtained , namely SRSF5, FGFR1, ACADVL, FOS, and LRP1. Their expression levels at 0, 3, 7, 10, 14, 21 and 28 days were quantified using RT–qPCR with 18S rRNA as the reference gene. Results demonstrated a consistent decrease in all five mRNAs over time. Mathematical models correlating mRNA levels with LPMI were established. Multi-index models exhibited superior fitting accuracy and predictive performance compared to single-index models, as validated using samples from 10, 18 and 25 days postmortem intervals, indicating greater practical applicability. This approach provides a new direction for forensic LPMI estimation research and contributes to the development of forensic pathology.

Project description:The estimation of the post-mortem interval (PMI) and the age-at-death (AAD) are crucial steps in the medico-legal investigation of unidentified human remains. Current methods to estimate PMI and AAD suffer from a lack of accuracy and objectivity, particularly when the remains are in an advanced state of decomposition. Recently, proteomics studies using animal models have identified potential biomarkers for PMI and AAD estimation. This study investigated the human bone proteome in four human body donors studied throughout decomposition outdoors. We compared different bone tissues (tibia and iliac crest) from body donors of known AAD, and compared bone samples taken shortly after death to bone samples upon complete skeletonization of the body. The effects of ageing phenomena (in vivo and post-mortem) and the surrounding environment on the variability and abundancy of the bone proteome were assessed.

Project description:In this study, we evaluated the effects of the post-mortem interval, defined as the number of days elapsed between the death of a donor and the isolation of hCECs, on the properties of hCECs. Using gene profiling, we compared the mRNA profiles of passage 1 hCECs (10 populations) cultivated in moculture with different post-mortem intervals and passage 2 hCECs (6 populations) with different post-mortem intervals from hTECs.
