Responses of various cells in the zebrafish olfactory epithelium after treatment with alarm substances

ABSTRACT: We performed single-cell transcriptome sequencing of the olfactory epithelium of AB strain zebrafish. We also applied alarm substances to one of the groups and studied the response of the olfactory epithelium to alarm substances by comparing the proportions of various cell types and transcriptional changes between the experimental and control groups.

A comparison of the intestinal phenotype and transcriptome profile analysis in the oral-fed mice with substances to affect the intestinal homeostasis

Project description:Epithelial cells in the intestinal mucosa maintain gut homeostasis by interacting with different types of microbiota. Proper appropriate immune responses in the intestinal epithelium are essential for the preservation of the intestinal homeostasis. In the present study, we aimed to identify genotypic and phenotypic changes in mice following oral feeding of various substances which has been shown to differentially affect intestinal homeostasis. We orally fed C57BL/6 mice for either one or seven days with one of the four substances: dextran sulfate sodium (DSS); Typhoid VI Polysaccharide vaccine (Vi vaccine); antibiotic cocktails (AB) of ampicillin, vancomycin, neomycin, and metronidazole; or(probiotics)consisting of Lactobacillus Rhamnosus R0011and L. Acidophilus R0052.While DSS and AB feeding resulted in severe gut pathology characterized by infiltration of inflammatory cells, epithelium shedding, and distortion of paneth cells. Vi vaccine and probiotics feeding resulted in phenotypic improvement of the gut health characterized by epithelial cell proliferation and increased formation of tight junctions between epithelial cells. Interestingly, microarray data showed significant increase in the expression levels of genes regulating cell proliferation and intestinal homeostasis in the gut epithelium of probiotics-and Vi vaccine-fed mice compared to DSS-or AB-fed mice. In addition, expression levels of genes regulating cell death and inflammation were significantly increased in the gut epithelium of DSS- and AB-fed mice. These results suggest that intestinal homeostasis play a pivotal role in maintaining gut health and, subsequently, in protecting host against enteric bacteria and external pathogens infection.

Inflammation and degeneration of the olfactory epithelium in an immuno-deficient zebrafish

Project description:Vertebrates are protected from pathogens by an adaptive and an innate immune system. In mammals, loss of the adaptive immune system severely compromises viability. Zebrafish, in contrast, can survive for relatively long without adaptive immunity, as demonstrated by the immuno-deficient Rag1 mutant. Here, we examine whether the innate immune system is activated in the mutant, and ask whether this has any effect on the nervous system, given the finding that inflammation is linked to neurodegeneration in higher vertebrates. Using microarray analysis of the olfactory epithelium, we show that innate immune responses are upregulated. Neuronal genes are down-regulated, and immunofluorescence for the neural marker HuC shows a progressive loss of olfactory sensory neurons in mutants. Propodium iodide uptake indicates a corresponding increase in cell death. Rag1 mutant fish progressively lose their sensitivity to an olfactory cue, the alarm pheromone Schreckstoff, indicating that neuro-degeneration has a behavioral significance. Taken together, these data establish that immuno-deficiency in the zebrafish is accompanied by neuro-inflammation and loss of neurons. The microarray data document transcriptional changes associated with activation of innate immunity in the context of immunodeficiency and also identify novel genes that are potentially related to function of the olfactory system. Olfactory transcriptomics in a mouse model of Alzheimer s disease

Project description:High-throughput sequencing of RNA from olfactory epithelium of mice modeling Alzheimer's disease with olfactory deficits. We sequenced cDNA libararies from the main olfactory epithelium (MOE) of nine mice from 3 strains (C57BL/6, T43 with P201S mutation, 129S5): 3 groups consists of 9 mice. RNA was extracted using a Qiagen RNAeasy kit and standard methods. All nine RNA samples were multiplexed together and sequenced in three lanes on the Illumina HiSeq platform. Project description:Expression profiling of mRNA abundance in the adult mouse olfactory epithelium during replacement of OSNs forced by the bilateral ablation of the olfactory bulbs. The experiment was done on 6 week old male C57Bl/6 mice. Olfactory epithelium tissue samples were collected on days 1, 5, and 7 after bulbectomy. The cellular processes activated by bulbectomy include apoptosis of mature olfactory sensory neurons, infiltration of macrophages and dendritic cells, stimulation of proliferation of basal cell progenitors, and differentation of new sensory neurons.

Whole-transcriptome profiling of regenerating olfactory epithelium following loss of CBX paralogs

Project description:The polycomb group protein, CBX8, is a neuron-specific component of Polycomb repressive complex 1 (PRC1) in the adult olfactory epithelium. We performed loss of function assays in an in vitro model of the regeneratng olfactory epithelium to determine the role of CBX8 in adult olfactory neurogenesis. Whole-transcriptome analysis highlighted the importance of CBX8-PRC1 in the regulation of adult neurogenesis in the olfactory epithelium.
