ScRNA of tumor associated lymphatic endothelial cells (LEC) from B16-OVA-VEGFC tumors implanted in Ifnγr1WT (Cre minus) and Ifnγr1iProx1 (Cre plus) mice

ABSTRACT: In order to determine transcriptional differences between lymphatic endothelial cell (LEC; CD45-CD31+pg38+) from B16-OVA-VEGFC melanoma tumors from Ifnγr1WT (Cre minus) and Ifnγr1iProx1 (Cre plus) mice, we sorted endothelial cells (DAPI-, CD31+, CD45-) from Ifnγr1WT (Cre minus) and Ifnγr1iProx1 (Cre plus) mice separately.

Bulk RNA sequencing of endothelial-derived and non-endothelial-derived BMSCs and OLCs of VE-Cadherin-Cre;LoxP-tdTomato mice

Project description:We performed lineage tracing experiments using VE-Cadherin-Cre;LoxP-tdTomato mice. In these mice, endothelial cells (ECs) and their progeny are permanently marked by tdTomato fluorescence. We found that a substantial subset of stromal cells is derived from ECs, as indicated by their tdTomato expression. These findings support the notion that endothelial to mesenchymal transition (EndoMT) contributes to hematopoietic bone marrow niche formation in mice. Here we sought to determine the transcriptomic differences between endothelial-derived (tdTomato-positive) and non-endothelial-derived (tdTomato-negative) bone marrow stromal cells (BMSCs) and osteo/chondrolineage progenitor cells (OLCs). Murine niche populations were obtained from collagenased bone fraction of VE-Cadherin-Cre;LoxP-tdTomato mice at 3 weeks (n=2) or 11 weeks (n=2) of age. BMSCs (CD45-TER119-CD31-CD144-SCA-1+ CD51+ cells) and OLCs (CD45-TER119-CD31-CD144-Sca1-CD51+ cells) were FACS-purified and sequenced.

Project description:Competitive interactions between emerging blood vessels determine the clonogenic contribution to developing vascualture. Using a multi-color cre reporter, CD31+CD45-VeCad+ clones were isolated and analyzed for expression differences. Tamoxifen labeled CD31+, CD45-VeCad+ tumor endothelial cells were isolated by FACS for analysis from separate tumors. The most frequent clone was compared to the least frequent clone between and across tumors. The strain of mice is C57BL/6.

Expression data from endothelial SP and MP cells

Project description:We identified an endothelial progenitor/stem like population in the endothelial fraction of preexsiting blood vessels. And we used microarray to see the difference of gene expression profiles between these two populations. Hindlimb muscle of wild type C57BL/6 were digested to single cells and stained with anti-CD31 antibody, anti- CD45 antibody, Propidium iodide and hoechst 33342. CD31+CD45- side population cells and CD31+CD45- main population cells were sorted by flow cytometry for RNA extraction and hybridized on Affimetrix microarrays.
