Project description:The role of eosinophils in the development and progression of malignant pleural effusion (MPE) remains unclear. However, MPE is a frequently occurring manifestation of advanced cancer and is often associated with a poor prognosis. While current treatment options for MPE can provide palliative care, alternative therapies are urgently needed. To investigate this role, eosinophil counts were determined using flow cytometry and immunofluorescence. Eosinophil-deficient mice (Eos-null) and Cd3δ -promoter Il5 transgenic (Il5 Tg) mice were subjected to a mouse model of MPE. Eosinophils were isolated by magnetic-activated cell sorting (MACS) and fluorescence-activated cell sorting (FACS), and their transcriptional profile was obtained by RNA sequencing. Finally, we evaluated the effects of the C-C motif chemokine ligand 11 (CCL11) and a C-C motif chemokine receptor 3 (CCR3) axis on MPE pathology using recombinant mouse CCL11 and a CCR3 inhibitor. In humans, eosinophil counts were increased in MPE and negatively correlated with the LENT score. In animal models, eosinophils were rapidly recruited to the pleural cavity when challenged with tumor cells. Interestingly, eosinophil deficiency worsened MPE, while an increase in eosinophils through various methods improved MPE. Furthermore, the recruitment of eosinophils may be influenced by the CCL11-CCR3 axis. Taken together, the study highlights the protective role of eosinophils against MPE formation, indicating their potential as a therapeutic strategy. Moreover, the CCL11-CCR3 axis may be a viable target for MPE treatment.

Project description:We hypothesised that eosinophil activation and gene expression were linked to their interaction with the activated stroma within the lymph node. However, the role of eosinophil stromal crosstalk and how this modulate eosinophil function is unknown. To directly assess this hypothesis, we activated lymph node derived stromal cells using an agonist antibody for LTR (Clone 4H8WH2). Considering eosinophils have long been associated with providing B cell proliferative and survival factors, we mimicked a cell-based activation model using purified naïve B cells given that B cell interactions with stroma can produce factors that can regulate eosinophil activation and migration. B cell-based activation as well as agonist antibody-based activation resulted in an activated stromal phenotype and were later used for co-culture with bone marrow-derived eosinophils. After 8 hours of co-culture with activated stroma, the eosinophils were purified, and bulk RNA sequencing was performed to identify global changes in their transcriptomes.

Project description:Skin is composed of diverse cell populations that cooperatively maintain homeostasis. Upregulation of the NF-kB pathway may lead to the development of chronic inflammatory disorders of the skin, but the early events in situ are not well understood. Through analysis of single cell RNA sequencing (scRNA-seq) data via iterative Random Forest Leave One Out Prediction (iRF-LOOP), an explainable artificial intelligence (X-AI) method, we identified an immunoregulatory role for a unique Prx1+ fibroblast subpopulation. Disruption of Ikkb/NFkB under homeostatic conditions in these fibroblasts paradoxically induced skin inflammation due to the over-expression of CCL11 (eotaxin-1) characterized by eosinophil infiltration and a subsequent Th2 immune response. Because the inflammatory phenotype resembled that seen in human atopic dermatitis (AD), we examined validated human AD skin samples and found that human AD fibroblasts also over-expressed CCL11 compared to control skin. Importantly, we demonstrate that monoclonal antibody treatment against CCL11 was therapeutically effective in reducing the eosinophilia and Th2 inflammation in vivo. Taken together, the murine model and human AD specimens point to dysregulated Prx1+ fibroblasts as a previously unrecognized etiologic factor that may contribute to the pathogenesis of AD-like lesions and suggest targeting CCL11 as a novel way to treat AD-like lesions.

Project description:The biocompatibility of immunoisolation devices is a major challenge for their use in cellular therapies. Utilisation of alginate for the encapsulation of pancreatic islets as a potential cellular therapy for type 1 diabetes has shown to be limited in terms of long-term graft survival, due to pericapsular fibrotic overgrowth. Fibrotic overgrowth is a complex process that involves several factors, including protein adsorption. This work investigates the protein adsorption profiles of plasma incubated alginate beads for the identification of key proteins that may play a role in the biocompatibility of this biomaterial.

Project description:Activation of inflammatory pathways in human IBD. Leukocyte recruitment pathways including those for eosiniphils are activated in the affected colon in IBD. However, the functional implications of this are not known. We hypothesized that pro-inflammatory eotaxin (CCL11) dependent networks would be up regulated in the colon of pediatric patients with Ulcerative Colitis (UC), and that these would regulate eosinophil recruitment to the gut. These experiments tested differential colon gene expression relative to these pathways in healthy and UC samples. Colon biopsy samples were obtained from UC patients at diagnosis, and healthy controls. The global pattern of gene expression was determined using GeneSpring software, and biological networks were identified using Ingenuity software. Data suggested that a leukocyte recruitment network which includeds CCL11 is up regulated in pediatric UC at diagnosis. The degree of up regulation of these genes compared to healthy controls was remarkably conserved within the UC patient group, suggesting common mechanisms of mucosal inflammation. These experiments tested differential colon gene expression relative to these pathways in healthy and UC samples. Keywords: Single time point in UC and healthy controls. Project description:While the function of many leukocytes in transplant biology has been well defined, the role of eosinophils is controversial and remains poorly explored. Conflicting data exist regarding eosinophils’ role in alloimmunity based on context and organ type. Due to their prevalence in the lung, and their defined role in other pulmonary pathology such as asthma, we set out to explore the role of eosinophils in the long-term maintenance of the lung allograft. We noted that depletion of eosinophils results in the generation of donor-specific antibodies. Eosinophil depletion increased memory B cell, plasma cell, and antibody secreting cell differentiation and resulted in de novo-generation of follicular germinal centers. Germinal center formation depended on the expansion of CD4+Foxp3-Bcl6+CXCR5+PD1+ T follicular helper cells (Tfh) which increase in number after eosinophil depletion. Mechanistically, we demonstrate that eosinophils prevent Tfh generation by acting as the dominant source of IFN-γ in an established lung allograft, thus facilitating Th1 rather than Tfh polarization of naïve CD4+ T cells. Our data thus describe a unique and previously unknown role for eosinophils in maintaining allograft tolerance and suggests that indiscriminate administration of eosinophil-lytic corticosteroids for treatment of acute cellular rejection may inadvertently promote humoral alloimmunity.
