A single-cell resolution spatial transcriptomics atlas of human multiple sclerosis lesion (MERSCOPE)

ABSTRACT: In this study, we conducted an in-depth investigation into the complex intercellular immune reactions in multiple sclerosis (MS). To accurately map the spatial transcriptomic landscape of MS, we designed a gene panel comprising 500 key genes and collected samples from the brain tissues of 14 patients with chronic active MS. Using MERFISH and 10x single-cell sequencing technologies, we successfully created detailed spatial transcriptional maps. Through comprehensive analysis of the data obtained, we identified several disease-specific glial cell subtypes associated with MS. Furthermore, we explored how these disease-related glial cells interact with peripheral immune cells, revealing their potential roles in the progression of the disease. This research not only provides new insights into the pathological mechanisms of multiple sclerosis but also offers significant scientific basis for the development of future therapeutic strategies.
