MeRIP-seq study of Vaccinia virus-infected Vero cell

ABSTRACT: We aimed to understand the dynamic interplay between host and virus by investigating transcriptomic and epitranscriptomic dynamics in the Vero E6 cells during Vaccinia Virus infection. In this study, we characterized N6-methyladenosine (m6A) modifications, pivotal regulators of transcript lifecycle, throughout three different post-infection time points using RNA-seq and m6A RNA immunoprecipitation sequencing (MeRIP-seq). We identified changes in host m6A epitranscriptome in the early phase of viral infection, which includes genes involved in host immunity, RNA biosynthesis, and protein ubiquitination. Leveraging temporal analysis of host transcriptomic and epitranscriptomic landscapes, this study offers a novel insight into the host-virus interaction, emphasizing the importance of RNA modifications in host transcripts as an early response to viral infection.
