Single cell sequencing of dura maters of dural metastasis models

ABSTRACT: Dural metastasis (DM) is a rarely researched disease with limited therapeutic options, largely due to our circumscribed understanding of dura mater. However, recently emerging studies suggest the dura mater is an immune hub and implements immunosurveillance of the central nervous system. These evidences imply potential immunotherapies of DM. Here, we established a preclinical murine model of DM. We have employed a single cell sequencing approach using 10x Genomics scRNA-seq to study the tumor microenvironment of DM model. In comparison, the dura maters of nontumor mice and leptomeningeal metastasis (LM) mice were used as control groups. Briefly, the dorsal dura maters are isolated from the head of nontumor mice, 4T1-LM mice and 4T1-DM mice on day 12 since tumor cell inoculation, and are enzymatically digested to single cells with collagenase VIII and DNase I.

RNA sequencing of meningeal lymphatic vessels in dural metastasis models

Project description:Dural metastasis (DM) is a rarely researched disease with limited therapeutic options, largely due to our circumscribed understanding of dura mater. However, recently emerging studies suggest the dura mater is an immune hub and implements immunosurveillance of the central nervous system. These evidences imply potential immunotherapies of DM. Here, we established a preclinical murine model of DM. We have employed a RNA sequencing approach using SMART-seq to study the meningeal lymphatic vessels of DM model. Briefly, on day 12, the dorsal dura maters were isolated from the head of 4T1-DM mice treated with PBS, ZOL or ZOL+Ang2, and were enzymatically digested to single cells with collagenase VIII and DNase I. The meningeal lymphatic endothelial cells (LECs) were harvested using fluorescent activated cell sorting, CD45-CD31+PDPN+ dural cells were sorted for sequencing.

Dynamic alterations of dural and bone marrow B cells in an animal model of progressive MS

Project description:In multiple sclerosis (MS), the leptomeninges (LM) are populated with immune cell aggregates that correlate with disease progression. The impact of LM inflammation on the adjacent dura is largely unknown. Using a mouse model of MS that induces brain LM inflammation and age-dependent disease progression, we found that encephalitogenic T cells and B220high B cells accumulate substantially in the brain LM and parenchyma of both young and aged mice, while the adjacent dura remains relatively inert. We also observed a population of anti-CD20-resistant B220low B cells in the dura and bone marrow that virtually disappear at disease onset and accumulate in the brain of young mice concomitant with disease remission. In contrast, aged mice show a paucity of brain-resident B220low B cells at the expense of class-switched B220high B cells accompanied by severe, chronic disease. In summary, dynamic changes in the brain, LM, and dural B cells are associated with age-dependent disease severity in an animal model of progressive MS.

Transcription profiling of mouse adult and juvenile dura mater (bone) to explore the reossification of large calvarial defects during development

Project description:BACKGROUND: Although reossification of large calvarial defects is possible in children, adults lack this tissue engineering capacity. In this study, the authors compared the differences in gene expression between juvenile and adult dura mater using a mouse cDNA microarray with 42,000 unique elements. METHODS: Non-suture-associated parietal bone was harvested from 6-day-old and 60-day-old mice. The dura mater was carefully dissected from the calvarial disk and snap-frozen. RNA was extracted from pooled dura mater for microarray analysis. The 25 most differentially expressed genes were listed, as were selected bone-related genes. In addition, quantitative real-time reverse-transcriptase polymerase chain reaction confirmation of selected genes-BMP-2, BMP-4, and BMP-7; and osteopontin (OP), osteocalcin (OC), and FGFR-1-was performed. RESULTS: Juvenile dura mater expressed significantly greater amounts of BMP-2 and OP. Minimal difference in OC expression was observed between juvenile and adult dura mater. Extracellular matrix proteins (Col3a1, 5a1, 6a1, and fibronectin 1), osteoblast differentiation markers (Runx2/Cbfa1, Itm2a, and FGFR-1), and the growth factor Ptn were among other genes with greater expression in juvenile dura mater. Markers of osteoclasts (Acp5, MMP9, Ctsk) and the multiple candidate gene Ntrk2 were also expressed at higher levels in the juvenile dura mater. CONCLUSIONS: These findings suggest a more differentiated osteoprogenitor population to exist along with a greater presence of osteoclasts in the juvenile dura mater relative to adults. In addition to establishing a baseline difference in gene expression between juvenile and adult dura mater, new genes potentially critical to the regenerative potential of juvenile calvaria were identified.

The dura mater acts as an autonomous B cell niche supporting immune responses during chronic Trypanosoma brucei infection

Project description:The dura mater hosts a rich population of B cell progenitors, yet its capacity to sustain B cell development during systemic lymphopenia remains unclear. Using Trypanosoma brucei infection, which induces peripheral B cell depletion, we demonstrate that the murine dura mater maintains intact B cell lymphopoiesis independently of bone marrow and spleen. We performed a comparative single cell transcriptomic profiling of the calvaria bone marrow, femur bone marrow, and dura mater from naive and Trypanosoma brucei-infected female mice. The sequencing rendered high quality data from the calvaria bone marrow and the dura mater, but the femur bone marrow failed to generated reliable data and was thus excluded from downstream analyses. Cerebral Venous Blood Flow Regulates Brain Fluid Clearance via Dural Lymphatics

Here, we examined the hypothesis that cerebral venous blood flow may participate in brain clearance as an intermediate compartment between the arterial and lymphatic poles of the vascular system.
