Single cell sequencing of dura maters of dural metastasis models


ABSTRACT: Dural metastasis (DM) is a rarely researched disease with limited therapeutic options, largely due to our circumscribed understanding of dura mater. However, recently emerging studies suggest the dura mater is an immune hub and implements immunosurveillance of the central nervous system. These evidences imply potential immunotherapies of DM. Here, we established a preclinical murine model of DM. We have employed a single cell sequencing approach using 10x Genomics scRNA-seq to study the tumor microenvironment of DM model. In comparison, the dura maters of nontumor mice and leptomeningeal metastasis (LM) mice were used as control groups. Briefly, the dorsal dura maters are isolated from the head of nontumor mice, 4T1-LM mice and 4T1-DM mice on day 12 since tumor cell inoculation, and are enzymatically digested to single cells with collagenase VIII and DNase I.
