Project description:We applied RNAseq analysis on microdissected cortical and medullary thick ascendind limb of Henle’s loop from WT and Cldn10 KO mice (cKO). Among 11369 genes sequenced 291 have an expression significantly modified (p<0.05) between MTAL and CTAL from WT mice. In cKO mice, among 12023 genes sequenced 359 were signicantly modified (p<0.05) between MTAL and CTAL. surprisingly, while 637 genes have significantly (p<0.05) different expression between MTAL WT and cKO only 66 were in this case between CTAL WT and cKO.

Transcriptional profiling of renal cells expressing wild type or mutant uromodulin isoforms.

Project description:Uromodulin is the most abundant urinary protein. It is exclusively produced and released in the urine by renal epithelial cells lining the thick ascending limb of Henle’s loop (TAL). Mutations in UMOD, the gene encoding uromodulin, cause autosomal dominant tubulointerstitial kidney disease uromodulin-related (ADTKD-UMOD). While the primary effect of all mutations, retention in the endoplasmic reticulum (ER), is well established, its downstream effects are still unknown. The aim of this study was to gain insight into ADTKD-UMOD pathogenesis through transcriptional profiling of immortalised mouse TAL cells (mTAL) transduced with lentiviral vectors encoding wild type or mutant (C150S) GFP-tagged human uromodulin.

Project description:To gain insight into the mechanisms underlying the defective autophagic activity and its functional impact on the pathogenesis of disease, we performed RNA sequencing (RNA-seq) of mRNA isolated from TAL cells purified from heterozyous Umod deletion mutation mice (DEL/+) and Umod +/+ littermates at 16 weeks, the full-blown stage of the disease. Our analysis revealed that 9566 genes were significantly differentially expressed between mutant and WT TAL cells, including 4706 upregulated and 4860 downregulated.

SMARCB1 regulates a TFCP2L1-MYC transcriptional switch promoting renal medullary carcinoma transformation and ferroptosis resistance. [ChIPseq and Cut&Tag]

Project description:Renal medullary carcinoma (RMC) is an aggressive tumour driven by bi-allelic loss of SMARCB1 and tightly associated with sickle cell trait. However, the cell-of-origin and oncogenic mechanism remain poorly understood. Using single-cell sequencing of human RMC, we defined transformation of thick ascending limb (TAL) cells into an epithelial-mesenchymal gradient of RMC cells associated with loss of renal epithelial transcription factors TFCP2L1, HOXB9 and MITF and gain of MYC and NFE2L2-associated oncogenic and ferroptosis resistance programs. We describe the molecular basis for this transcriptional switch that is reversed by SMARCB1 re-expression repressing the oncogenic and ferroptosis resistance programs leading to ferroptotic cell death. Ferroptosis resistance links TAL cell survival with the high extracellular medullar iron concentrations associated with sickle cell trait, an environment propitious to the mutagenic events associated with RMC development. This unique environment may explain why RMC is the only SMARCB1-deficient tumour arising from epithelial cells, differentiating RMC from rhabdoid tumours arising from neural crest cells.
