Kaposi’s Sarcoma-associated Herpesvirus Confers Mesenchymal Stem Cells Tumorigenicity: A Cell and Animal Model of Virally Associated Osteosarcoma

ABSTRACT: This study explores the potential etiological role of KSHV in osteosarcoma development. Given that osteosarcoma originates from mesenchymal stem cells (MSCs), we infected human bone marrow MSCs with KSHV. The infected MSCs demonstrated enhanced osteogenic differentiation and acquired increased tumorigenic potential. When these KSHV-infected MSCs were injected near the femur of BALB/c nude mice, tumors developed on the bone surface, displaying a "Codman triangle" periosteal reaction and histological features characteristic of high-grade sarcoma—as well as expression of osteosarcoma markers STAB2 and MDM2. Transcriptomic analysis comparing xenograft tumors with human KSHV-associated osteosarcomas revealed a similar pattern of signaling pathway activation, including the EEF1A2-mTOR axis and PDPN.
