ABSTRACT: Winged bean (WB), Psophocarpus tetragonolobus, is a tropical legume, the potential of which is not yet been understood. We found that a 5 week-oral administration of WB seed extract inhibited wrinkle formation induced by repeated tape stripping (TS), which is a model of lichenification in human chronic eczematous dermatitis. To elucidate mechanism of the effect of WB on this model, we applied microarray analysis.

We found that a 5 week-oral administration of WB seed extract inhibited wrinkle formation induced by repeated tape stripping (TS), which is a model of lichenification in human chronic eczematous dermatitis. To elucidate mechanism of the effect of WB on this model, we applied microarray analysis. Skin barrier was disrupted by repeated application and removal (tape stripping; TS) of cellophane tape on the dorsal skin of the left side. This procedure was started after 1 week-administration of winged bean (WB) extract, and was conducted 3 times per week for 4 weeks. Dorsal skin samples from three experimental groups were used for microarray experiment. These groups were the non-TS (NT), TS, and TS with oral administration of WB extract (TS/WB). The DNA microarray experiment was performed using Affymetrix Mouse Genome 430 2.0 Array.

The stratum corneum transcriptome in atopic dermatitis can be assessed by tape stripping

Project description:Background: Skin biopsies represent a gold standard in skin immunology and pathology but can cause pain and induce scarring. Non-invasive techniques will facilitate study recruitment of e.g. patients with pediatric atopic dermatitis (AD), hand eczema or facial dermatitis. Objective: By RNA sequencing, we examined whether the stratum corneum transcriptome in AD skin can be assessed by tape stripping, as compared to the epidermal transcriptome of AD in skin biopsies. To make the procedure clinically relevant tape strips were stored and shipped at room temperature for up to 3 days. Methods: Nine adult Caucasian AD patients and three healthy volunteers were included. Tape samples were collected from non-lesional and lesional skin. Biopsies were collected from lesional skin and were split into epidermis and dermis. Total RNA was extracted, and shotgun sequencing was performed. Results: Shotgun sequencing could be performed on skin cells obtained from two consecutive tape strips which had been stored and shipped at room temperature for up to three days. The most prominent differences between the tape strip and biopsy derived transcriptome were due to structural genes, while established molecular markers of AD, including CCL17, CCL22, IL17A and S100A7-S100A9, were also identified in tape strip samples. Furthermore, the tape strip derived transcriptome showed promise in also analysing the skin microbiome. Conclusion: Our study shows that the stratum corneum (SC) transcriptome of AD can be assessed by tape stripping the skin, supporting that this method may be central in future skin biomarker research.

IL22RA2 is a Smad7 target mediating alleviation of dermatitis and psoriatic phenotypes in mice

Project description:Long-term management of inflammatory skin diseases is challenging because of side effects from repeated use of systemic treatments or topical corticosteroids. This study sought to identify novel mechanisms and developmental therapeutics for these diseases using genetic models and pharmacological approaches. We found mice overexpressing Smad7 in keratinocytes, but not mice overexpressing the N-terminal domain of Smad7 (N-Smad7), were resistant to imiquimod (IMQ)-induced Th1/17 and Th2 type inflammation. We generated a truncated Smad7 protein encompassing C-terminal Smad7 and PY motif fused with cell-penetrating Tat peptide (Tat-PYC-Smad7). Topically applied Tat-PYC-Smad7 to inflamed skin entered cells upon contact and attenuated IMQ-, 2,4-dinitrofluorobenzene-induced atopic dermatitis (AD) and tape-stripping inflammation (TS). RNAseq analyses of mouse skin exposed to these insults identified that in addition to inhibiting TGFβ/NFκB, Smad7 blunted IL22/STAT3 activation and associated pathogenesis, which is due to Smad7 transcriptionally upregulating IL22 antagonist IL22RA2. Mechanistically, Smad7 facilitated nuclear translocation and DNA binding of C/EBPβ to IL22RA2 promoter for IL22RA2 transactivation. Consistent to the above observations in mice, transcript levels of IL22RA2 were increased in human atopic dermatitis and psoriasis lesions with clinical remission. Our study identified the anti-inflammation functional domain of Smad7 and suggests mechanism and feasibility for developing Smad7-based biologics as a topical therapy for skin inflammatory disorders.

The proteome of hand eczema assessed by tape stripping

Project description:Hand eczema (HE) is a prevalent skin disease. However, classification of HE into different subtypes remains challenging. Limited number of prior studies have employed invasive biopsy-based strategies; yet, studies of the HE proteome using non-invasive tape stripping methodology have not been reported. In this study, we wanted to assess whether global proteomic analysis of skin tape strip samples can be used for sub-classification of HE patients. Tape strips were collected from patients with HE and healthy skin. Liquid chromatography–mass spectrometry (LC/MS) proteomics was performed, and the global protein expression was analyzed. We identified 2,919 proteins in stratum corneum-derived skin cells from tape strip samples. Compared to healthy skin, the lesional samples from HE patients exhibited increased expression of immune-related markers and a decreased expression of structural barrier proteins. The difference between HE subtypes was restricted to the lesional skin areas, and included an increased expression of skin barrier-related proteins independently of the concurrent AD. In conclusion we found, that the non-invasive tape strip method used in combination with LC/MS proteomics can be used for analysis of skin protein expression in HE patients. Thus, the method shows potential for assessing the proteomic differences between subtypes of HE, and biomarker discovery.
