Project description:Dietary methionine restriction (MR) has been shown to increase lifespan and decrease adiposity in rodents. This study was designed to examine the transcriptional effects of MR in metabolically relevant tissues. This experiment contains data from the liver. We analyzed MR-induced changes in gene expression using pooled RNA from liver of rats fed either a control purified amino acid diet (DL-methionine content of 0.86%) (CON) or a methionine-restricted diet (DL-methionine content of 0.172%)(MR). Rats were fed Purina rodent diet 5001 until 32 days of age and were then randomly assigned to be fed CON diet or MR diet for 20 months.

The effect of Methionine Restriction using oxidized casein as protein source on the transcriptome of mouse IWAT

Project description:Ingestion of a diet with a restricted Methionine (MR) content results in robust activation of a set of browning genes in IWAT (inguinal white adipose tissue). So far, data were obtained using a mix of elemental amino acids as the dietary protein source, which was replaced in this experiment with casein. To reduce methionine content, casein was oxidized and methionine was added back to a concentration similar to the diets using elemental amino acids as protein source. Results show that mice reduce body weight and adiposity on MR containing oxidized casein similar to MR containing elemental amino acids. Similarly, energy expenditure, as well as food and water intake were increased. The RNAseq experiment was conducted to compare the effect of MR using these two protein sources on the IWAT transcriptome.

The effect of dietary methionine restriction using different protein sources on the transcriptome of mouse livers

Project description:Dietary Methionine restriction (MR) has been demonstrated to induce a set of hepatic responses, including activation of hepatic stress response and suppression of hepatic lipogenesis. In the current study, we used two different protein sources to achieve MR. Casein was used in normal protein (NP, 20% casein) and low protein (LP, 5% casein) diet. Soy protein was used in 20Soy (20% soy), 20Soy+SAA (20% soy, Met and Cys was added back to match their concentration in NP diet), and 10Soy (11.5% soy, the Met concentration matches LP diet). Results show that mice that were fed LP and 10Soy diet have lower body weight and adiposity and increased food and water intake, energy expenditure, and serum FGF21 levels compared to their relative controls. The RNAseq experiment was conducted to compare the effect of MR using these two protein sources on the hepatic transcriptome.

The role of NRF2 in the effects of methionine restriction on the transcriptome of mouse livers

Project description:Dietary methionine restriction results in a robust activation of transcription factor, NRF2, and its target gene expression in the liver. In the current study, we observed liver-specific knockout NRF2 did not affect the physiological responses due to MR consumption. Therefore, there are other transcription factors compensate for the loss of NRF2 in the liver to mediate the effect of MR. The RNAseq experiment was comducted to compare the effect of MR diet in NRF2 WT and KO mice on hepatic transcriptome.

Dietary methionine restriction impairs anti-tumor immunity through gut microbiota

Project description:Dietary methionine restriction represses growth and improves therapeutic responses in several pre-clinical settings. However, how this dietary intervention impacts cancer progression in the context of the immune system is unknown. Here we analyzed the CD45+ immune cells from the small intestine of control (CTRL) diet or methionine-restricted (MR) diet fed tumor-free C57BL/6J donor mice and tumor-bearing Apc <min+/-> recipient mice transplanated with feces from these diet-fed tumor-free C57BL/6J mice by scRNA-seq. Our analysis indicate that fecal microbes from methionine-restricted tumor-free C57BL/6J mice are sufficient to represss T cell activation in the small intestine of Apc <min+/-> mice.

Methionine restriction alleviate cognitive dysfunction in Alzheimer’s disease: the mediating roles of gut microbiota-brain axis

Project description:We reported that a 4-month methionine-restricted diet (MR) intervention reduced the cognitive impairment in Alzheimer’s disease. Here, we introduce the transcriptomics data of the mouse hippocampus to further study how MR regulates important biological processes and pathways. After mapping clean RNA-seq reads of all mice against the Mus musculus genome, we detected 56,412 genes (including 12,527 new predicted genes) with fragments per kilobase of transcripts per million mapped reads (FPKM) value. Gene set enrichment analysis (GSEA) was used for pathway or functional analysis of RNA-seq data.

Methionine metabolism shapes T helper cell responses through regulation of epigenetic reprogramming [ChIP-Seq]

Project description:Epigenetic modifications on DNA and histones regulate gene expression by modulating chromatin accessibility to transcription machinery. Chromatin-modifying enzymes are dependent on metabolic intermediates for chromatin remodeling, linking nutrient availability and cellular metabolism to the cellular epigenetic landscape. Here we identify methionine as a key nutrient affecting T cell epigenetic reprogramming in CD4+ T helper (Th) cells. Using metabolomic approaches, we showed that methionine is rapidly taken up by activated T cells and then serves as the major substrate for the biosynthesis of S-adenosyl-L-methionine (SAM), the universal methyl donor for cellular methyltransferases. Conversely, methionine restriction (MR) depletes intracellular SAM pools, reduces global histone H3K4 methylation (H3K4me3) in T cells, and reduces H3K4me3 levels at the promoter regions of key genes involved in CD4+ Th17 cell proliferation and cytokine production. Applied to the mouse model of multiple sclerosis (experimental autoimmune encephalomyelitis), dietary methionine restriction reduced the expansion of pathogenic Th17 cells in vivo, leading to reduced T cell-mediated neuroinflammation and disease onset. Overall our data identify methionine as a key nutritional factor that shapes T cell proliferation, differentiation, and function in part through regulation of histone methylation in T cells.
