Spatial transcriptomics reveals the immunomodulatory activities of an oncolytic adenovirus armed with a type I interferon in a replication-permissive immunocompetent pancreatic cancer model

ABSTRACT: Oncolytic adenoviruses (OAds) are emerging as a promising class of viro-immunotherapy. Here, we focused on a replication-competent OAd armed with a type I interferon (OAd-IFN) and assessed its immunological activities in a replication-permissive, immunocompetent Syrian hamster pancreatic cancer model. Our spatial transcriptomic analysis revealed the critical role of locally expressed type I IFN in the immunomodulatory activities of OAd-IFN in the tumor microenvironment.

Project description:In this study we demonstrate the effect of oncolytic adenoviruses armed with CXCL9, CXCL10 and IL-15 to infect cancer cells, secrete the encoded protein and drive gradient-dependent T-cell attraction. Our in vivo validation demonstrated an increased intratumoral CD4+ and CD8+ T-cell infiltration following treatment with armed viruses compared to control groups. Finally, RCC cells were screened for tumor-specific peptides to validate their immunogenicity in a (personalized) oncolytic cancer vaccine approach, previously referred to as PeptiCRAd.

In vivo armed macrophages curb liver metastasis through tumor reactive T cell rejuvenation (TAD18)

Project description:Despite recent progress in cancer treatment, liver metastases persist as an unmet clinical need. This is partly attributable to the immunosuppressive microenvironment of liver metastases, which inhibits immune activation. To unleash anti-tumoral immune responses against liver metastases, we armed liver and tumor-associated macrophages to coordinately express tumor antigens (TAs), IFNα and IL-12. The combination of TAs with both cytokines yielded the most potent therapeutic response, resulting in the eradication of established liver metastases in several mouse models of the disease. Mechanistically, armed macrophages expanded tumor reactive CD8+ T cells, which in the presence of IFNα and IL-12 acquired features of progenitor exhausted T cells and killed cancer cells independently of CD4+ T cell help. IFNα and IL-12 produced within the liver tissue by armed macrophages rewired cell interactions within the tumor microenvironment and reprogrammed antigen presenting cells to further support T cell effector functions. Tumor reactive T cell rejuvenation and strong therapeutic activity were observed in distinct liver metastasis mouse models of colorectal cancer and melanoma, expressing either surrogate tumor antigens, naturally occurring private neoantigens or tumor-associated antigens. Altogether, our findings support the translational potential of in vivo armed liver macrophages expressing TA and immune-activating cytokines to expand and rejuvenate tumor reactive T cells for the treatment of liver metastases.

Sen1 and Rrm3 ensure permissive topological conditions for replication termination [ChIP-seq]

Project description:Replication forks terminate at TERs and telomeres. Forks that converge or encounter transcription generate topological stress. Combining genetic, genomic and imaging approaches we found that Rrm3hPif1 and Sen1hSenataxin helicases assist termination at TERs, Sen1 at telomeres. rrm3 and sen1 are synthetic lethal, fail to terminate replication exhibiting lagging chromosomes and fragility at TERs and telomeres. sen1 rrm3 build up RNA-DNA hybrids at TERs, sen1 accumulates RNPII at TERs and telomeres. Double mutants exhibit X-shaped gapped or reversed converging forks. Rrm3 and Sen1 restrain Top1 and Top2 activities, preventing toxic accumulation of positive supercoil at TERs and telomeres. We suggest that Rrm3 and Sen1 coordinate the activities of fork-associated Top1 and Top2 with those of gene loop-associated Top1 and Top2 by preventing DNA and RNA polymerases slowing down when forks encounter transcription head-on or codirectionally, respectively. Hence Rrm3 and Sen1 are essential to generate permissive topological conditions for replication termination.
