ABSTRACT: Obesity is a chronic metabolic disorder that increases the risk of various metabolic diseases, such as type 2 diabetes and metabolic dysfunction-associated steatohepatitis (MASH). While previous studies have shown that Five Substances Powder with Poria can alters gut microbiota to improve glycolipid metabolism and obesity, the underlying mechanisms still need to be clarified. The present study aimed to explore the potential mechanisms of Five Substances Powder with Poria in suppressing obesity more deeply through an integrated research strategy.

Gene expression in mouse liver in response to krill powder

Project description:The effect of krill powder, a mixed source of protein and n-3 PUFAs from Antarctic Krill (Euphausia superba), on hepatic gene expression was analyzed in CBA/J mice. Mice were fed a low-fat control diet or a 3% (w/w) krill powder low-fat diet for three months. Gene expression profiling on liver samples revealed that the krill powder supplemented diet modulated a large number of pathways compared to the control diet, and we focused on the genes involved in metabolic processes. Pathways involved β-oxidation, glucose metabolism, and amino acid catabolism were downregulated. In contrast, genes involved in the mitochondrial electron transport chain were upregulated. Thus, a krill powder supplemented diet had potent and specific effects on energy metabolism and oxidative phosphorylation at the gene level. This indicates that krill powder supplementation could be an approach to prevent age-related decline in mitochondrial respiratory chain function and weight loss.

Differential expression of Transcriptomics and Metabolomics in different developmental periods of poria cocos

Project description:<p>Poria cocos, a traditional medicinal fungus, exhibits a wide array of biological activities. Through the integration of metabolomic and transcriptomic analyses, we have determined that the development of Poria cocos mycelia is predominantly propelled by biochemical reactions triggered by oxidative stress. Within this process, the mitogenactivated protein kinase (MAPK) signaling pathway assumes a pivotal regulatory function. As Poria cocos reaches its maturation stage, we have identified several functional substances, such as oleanolic acid, 18β-glycyrrhetinic acid, hawthorn acid, ergosterol and dehydroergosterol, among others. Our findings have unveiled the molecular mechanism underlying the formation of sclerotia in Poria cocos, providing crucial insights into its biological traits and its response to environmental stressors. These functional substances underscore the potential of Poria cocos in the development and application of nutraceuticals.</p>

Exo-proteomics analysis in the microbial antagonist Pythium oligandrum cultured with inactivated oomycete mycelial powder

Project description:Pythium oligandrum is an important microbial antagonist and biocontrol agent. P. oligandrum protease activity was found to be induced when cultured with inactivated oomycete mycelial powder from one of its hosts/prey Pythium myriotylum. The dataset consists of exo-proteomics analysis of culture supernatants from 2 d P. oligandrum GAQ1 strain shake-flask cultures with Plich medium supplemented with either glucose (samples GAQ1G-L4 to 6) or inactivated mycelial powder (samples GAQ1P-L7 to 9). A total of 453 proteins or protein groups were detected in at least one of the culture supernatants, each with at least two unique peptides. There were 91 proteins more abundant or exclusively detected in the mycelial powder cultures, and 96 proteins more abundant or exclusively detected in the glucose cultures. Proteases were among the most common annotations among proteins more abundant in mycelial powder cultures. The dataset highlights proteins potentially involved in P. oligandrum antagonism and in the utilization of carbon and nitrogen sources.
