ABSTRACT: Purpose: This study investigates the impact of NRG1 on the cardiac adaptation to pressure overload and analyzes the underlying mechanisms. Methods: Male C57BL/6NRj mice underwent sham or transverse aortic constriction (TAC) surgery, received minipumps with either saline or NRG1 and were followed by echocardiography and BRB-seq analysis. Results: NRG1-induced functional improvement of the heart after TAC.

Characteristics of myocardial structure and central carbon metabolism during the early and compensatory stages of cardiac hypertrophy

Project description:Cardiac hypertrophy is a classical forerunner of heart failure and myocardial structural and metabolic remodeling are closely associated with cardiac hypertrophy. We aim to investigate the characteristics of myocardial structure and central carbon metabolism of cardiac hypertrophy at different stages. Using echocardiography and pathological staining, early and compensatory cardiac hypertrophy were respectively defined as within 7 days and from 7 to 14 days after transverse aortic constriction (TAC) in mice. Among mass-spectrometry-based metabolomics, we identified 45 central carbon metabolites. Differential metabolite analysis showed that six metabolites, including citrate, cis-aconitate and so on, decreased significantly on day 1 after TAC. Ten metabolites, including L-lactate, (S)-2-hydroxyglutarate and so on, were obviously changed on days 10 and 14. Pathway analysis showed that these metabolites were involved in seven metabolic pathways, including carbohydrates, amino acids and so on. Western blot showed the expression of ATP-citrate lyase, malate dehydrogenase 1 and lactate dehydrogenase A in myocardium changed markedly on day 3, while the phosphorylation level of AMP-activated protein kinase did not show significantly difference. Our study reveals the characteristics of myocardial structure and central carbon metabolism of cardiac hypertrophy at different stages, which may have a suggestive role in the early diagnosis of cardiac hypertrophy.

Neuregulin 1 type III reduces severity in a mouse model of Congenital Hypomyelination Neuropathy

Project description:Purpose: We hypothesised that neuregulin 1 type III would reverse CHN. Using a faithful mouse model of CHN, we rescued the myelination defects by activation of Nrg1 type III. Unexpectedly, rescue did not depend on upregulation of Egr2 or major myelin genes. Therefore we asked whether we could identify any transcriptomic signature of the canonical pathways downstream of Nrg1 type III that could explain the increase in myelination in the sciatic nerve. Methods: Sciatic nerve mRNA profiles of 30-day-old wild-type (WT), P0 mutant (Q215X/+), trangenic HA-NRG1 type III, and HA-NRG1 type III:Q215X mice were generated by deep sequencing, in duplicate, using Illumina HiSeq2500 . The sequence reads that passed quality filters were aligned to the mouse reference genome (mm10) with the transcript annotation (UCSC mm10 annotation download from TopHat website) using TopHat (v2.0.9). Quantification for the gene expression and differential expression analysis were done by the software Cufflinks (v2.2.0). Results: A comprehensive transcriptomic analysis of signals downstream of neuregulin 1 type III in rescued CHN nerves revealed imbalanced activation of MAPK/ERK signals and minor myelin genes, such as Pmp2 and Omg. Conclusions: Our findings demonstrate that although augmented Nrg1 type III activity modifies the composition of myelin, it still promotes thicker myelin and ameliorates nerve conduction velocity in a mouse model of CHN. Controlled increase of Nrg1 type III activity may be a therapeutic strategy in some hypomyelinating neuropathies.

The Role of Endothelial Autocrine NRG1/ERBB4 Signaling in Cardiac Remodeling

Project description:Neuregulin-1 (NRG1) is a paracrine growth factor, secreted by cardiac endothelial cells (Ecs) in conditions of cardiac overload/injury. The current concept is that the cardiac effects of NRG1 are mediated by activation of ERBB4/ERBB2 receptors on cardiomyocytes. However, recent studies have shown that paracrine effects of NRG1 on fibroblasts and macrophages are equally important. Here, we hypothesize that NRG1 autocrine signaling plays a role in cardiac remodeling. We generated EC–specific Erbb4 knockout mice to eliminate endothelial autocrine ERBB4 signaling without affecting paracrine NRG1/ERBB4 signaling in the heart. We first observed no basal cardiac phenotype in these mice up to 32 weeks. We next studied these mice following transverse aortic constriction (TAC), exposure to angiotensin II (Ang II) or myocardial infarction in terms of cardiac performance, myocardial hypertrophy, myocardial fibrosis and capillary density. In general, no major differences between EC–specific Erbb4 knockout mice and control littermates were observed. However, 8 weeks following TAC both myocardial hypertrophy and fibrosis were attenuated by EC–specific Erbb4 deletion, albeit these responses were normalized after 20 weeks. Similarly, 4 weeks after Ang II treatment myocardial fibrosis was less pronounced compared to control littermates. These observations were supported by RNA-sequencing experiments on cultured endothelial cells showing that NRG1 controls the expression of various hypertrophic and fibrotic pathways. Overall, this study shows a role of endothelial autocrine NRG1/ERBB4 signaling in the modulation of hypertrophic and fibrotic responses during early cardiac remodeling. This study contributes to understanding the spatio-temporal heterogeneity of myocardial autocrine and paracrine responses following cardiac injury.

Neuregulin-1β regulation of embryonic endothelial progenitor cell survival.

Project description:Endothelial progenitor cells (EPCs) are mobilized into the vascular space and home to damaged tissues, where they promote repair in part through a process of angiogenesis. Neuregulins (NRGs) are ligands in the epidermal growth factor family that signal through type I receptor tyrosine kinases in the erbB family (erbB2, erbB3, and erbB4) and regulate endothelial cell biology, promoting angiogenesis. Stimuli such as ischemia and exercise that promote EPC mobilization also induce cleavage and release of transmembrane NRG from cardiac microvascular endothelial cells (CMECs). We hypothesized that NRG/erbB signaling may regulate EPC biology. Using an embryonic (e)EPC cell line that homes to and repairs injured myocardium, we were able to detect erbB2 and erbB3 transcripts. Identical receptor expression was found in EPCs isolated from rat bone marrow and human whole blood. NRG treatment of eEPCs induces phosphorylation of kinases including Akt, GSK-3β, and Erk1/2 and the nuclear accumulation and transcriptional activation of β-catenin. NRG does not induce eEPC proliferation or migration but does protect eEPCs against serum deprivation-induced apoptosis. These results suggest a role for tissue-derived NRG in the regulation of EPC survival.

Load-Dependent Changes in Left Ventricular Gene Expression in a Pathophysiologically Relevant Murine Model of Reversible Heart Failure (2 weeks heart failure, 2 weeks reversal)

Project description:Wild type, female, C57BL/6 mice were subjected to sham (n=6) surgery, or TAC + MI to cause progressive LV remodeling (n=13). At 2wks post-TAC, one group of mice underwent de-banding (HF-DB, n=6), whereas in a second group of mice the band remained intact (HF_shDB; n = 7). LV remodeling was evaluated by 2D echocardiography at 14 days post-TAC+MI , and 2 wks post-debanding. At 4 wks the hearts were excised and analyzed for changes in gene expression using transcriptional profiling.

Neuregulin-1 prevents death from a normally lethal respiratory viral infection.

Project description:Respiratory infections with RNA viruses such as respiratory syncytial virus (RSV) and influenza lead to significant morbidity and mortality. Using a natural rodent pathogen, Sendai virus (SeV), which is similar to RSV, mice made atopic with house dust mite survived a normally lethal SeV infection. Since we previously demonstrated a critical role for CD11c+ cells in the immune axis linking SeV to post-viral airway disease we posited that some of the protection against lethal SeV infection could be due to CD11c+ cells from atopic mice. Therefore, we performed RNA-seq analysis of CD11c+ cells isolated from atopic and non-atopic (NA) mouse lungs. While there were several dysregulated genes, one gene product whose expression was increased several-fold in atopic CD11c+ cells was neuregulin-1(NRG1) whose protein was found to be markedly elevated in the lungs and bronchoalveolar lavage fluid of atopic mice. When NRG1 was administered to naïve (non-atopic) mice it protected them from death with both SeV and mouse adapted influenza A virus (IAV). Survival was associated with reduced alveolar epithelium permeability and reduced phosphorylation of mixed lineage kinase domain-like (MLKL) protein indicating inhibition of necroptosis. In conclusion, our data demonstrate a unique function of NRG1 in respiratory viral infections by reducing alveolar leak, inhibiting epithelial necroptosis, and promoting homeostatic regulation of airway epithelium, all of which associate with markedly reduced mortality to the respiratory viral insult.

Neuregulin-1β Improves Uremic Cardiomyopathy and Renal Dysfunction in Rats.

Project description:Chronic kidney disease is a global health problem affecting 10% to 12% of the population. Uremic cardiomyopathy is often characterized by left ventricular hypertrophy, fibrosis, and diastolic dysfunction. Dysregulation of neuregulin-1β signaling in the heart is a known contributor to heart failure. The systemically administered recombinant human neuregulin-1β for 10 days in our 5/6 nephrectomy-induced model of chronic kidney disease alleviated the progression of uremic cardiomyopathy and kidney dysfunction in type 4 cardiorenal syndrome. The currently presented positive preclinical data warrant clinical studies to confirm the beneficial effects of recombinant human neuregulin-1β in patients with chronic kidney disease.
