Project description:Nutrient availability, in particular that of carbon (C) and nitrogen (N), is one of the most important factors for the regulation of plant metabolism and development. In addition to independent utilization, the ratio of C to N metabolites in the cell is also important for the regulation of plant growth including. Plants sense and respond to the balance of carbon (C) and nitrogen (N) nutrients (C/N-nutrient) available to them, a process called the C/N-nutrient response. We previously demonstrated that disrupted high C/low N stress condition promotes the senescence progression in Arabidopsis plants. However, the molecular basis of C/N-nutrient responsive senescence regulation remains unclear. In this study, we carried out proteome analysis of phosphorylation dynamics in response to high C/low N nutrient stress.

Project description:Post-germination, emerging seedlings rely on storage reserves to fuel cellular metabolism until photoautotrophic growth is established. Seed reserves can also sustain growth when seedlings are occluded from light, by for instance, soil or debris, which delays the switch to photosynthesis-driven growth. This study has shown that HEXOKINASE 1 (HXK1) supports early seedling growth, particularly during periods of prolonged darkness or low light. HXK1 is an evolutionarily conserved enzyme that catalyzes the first glycolytic step in cellular respiration for energy production. We show that HXK1 also has a broader function in nutrient resource management, balancing the transcriptional control of energy generating vs demanding processes with the seedling internal carbon status.

Project description:Immune responses in plants are triggered in part by conserved microbe-associated molecular pattern (MAMP) molecules such as bacterial flagellin. Upon MAMP perception, plants rapidly turn on the induction of numerous defense-related genes. We have identified a novel type of plant innate immunity elicitor, protease IV from Pseudomonas aeruginosa. Genome-wide transcriptomic profiles obtained with Affymetrix Arabidopsis ATH1 GeneChips® of 10-day old Arabidopsis seedlings treated with 20 nM purified protease IV for 1 hour were compared to published bacterial flagellin- and oligogalacturonide-triggered responses. We used microarrays to characterize the global transcriptomic changes in Arabidopsis seedlings upon protease IV treatment.

Project description:Comparison of TopHat alignments and assessment of spurious splice junctions for 32nt and 76nt read lengths. Total RNA from 2-week-old Arabidopsis thaliana (ecotype Columbia) seedlings grown on MS plates was isolated using RNeasy Plant Mini Kit from Qiagen. To remove any contaminating DNA, RNA was treated with DNAse. Isolation of poly (A) mRNA and preparation of cDNA library were carried out using the Illumina TrueSeq RNA kit. Sequencing (72 cycle) was done on Illumina Genome Analyzer II.
