DNA Methyltransferase M.PaeTBCFI in a chronically adapted Pseudomonas aeruginosa strain

Project description:Pseudomonas aeruginosa is an opportunistic pathogen which causes acute and chronic infections that are difficult to treat. Comparative genomic analysis has showed a great genome diversity among P. aeruginosa clinical strains and revealed important regulatory traits during chronic adaptation. While current investigation of epigenetics of P. aeruginosa is still lacking, understanding the epigenetic regulation may provide biomarkers for diagnosis and reveal important regulatory mechanisms. The present study focused on characterization of DNA methyltransferases (MTases) in a chronically adapted P. aeruginosa clinical strain TBCF10839. Single-molecule real-time sequencing (SMRT-seq) was used to characterize the methylome of TBCF. RCCANNNNNNNTGAR and TRGANNNNNNTGC were identified as target motifs of DNA MTases, M.PaeTBCFI and M.PaeTBCFII, respectively.

In-Vivo Expression Profiling of Pseudomonas aeruginosa Infections Reveals Niche-Specific and Strain-Independent Transcriptional Programs

Project description:Pseudomonas aeruginosa is a threatening, opportunistic pathogen causing disease in immunocompromised individuals. The hallmark of P. aeruginosa virulence is its multi-factorial and combinatorial nature. It renders such bacteria infectious for many organisms and it is often resistant to antibiotics. To gain insights into the physiology of P. aeruginosa during infection, we assessed the transcriptional programs of three different P. aeruginosa strains directly after isolation from burn wounds of humans. We compared the programs to those of the same strains using two infection models: a plant model, which consisted of the infection of the midrib of lettuce leaves, and a murine tumor model, which was obtained by infection of mice with an induced tumor in the abdomen. All control conditions of P. aeruginosa cells growing in suspension and as a biofilm were added to the analysis. We found that these different P. aeruginosa strains express a pool of distinct genetic traits that are activated under particular infection conditions regardless of their genetic variability. The knowledge herein generated will advance our understanding of P. aeruginosa virulence and provide valuable cues for the definition of prospective targets to develop novel intervention strategies.

Gene expression in AES-2 clonal isolates of Pseudomonas aeruginosa

Project description:Pseudomonas aeruginosa (P. aeruginosa) lung infection is a significant cause of mortality in patients with cystic fibrosis (CF). Most CF patients acquire unique P. aeruginosa strains from the environment; however clonal strains have been identified in CF communities in several countries. Two clonal strains infect 10% to 40% of patients in three CF clinics in mainland eastern Australia. The expression profiles of four planktonically-grown isolates of one Australian clonal strain (AES-2), and four non–clonal CF P. aeruginosa isolates were compared to each other and to the reference strain PAO1 using the Affymetrix P. aeruginosa PAO1 genome array, to gain insight into properties mediating the enhanced infectivity of AES-1. The isolates were subsequently grown as 3-day old biofilms and similarly extracted for RNA and compared as above. Data analysis was carried out using BIOCONDUCTOR software. A549 lung pneumocytes cell line infected with wildtype and deletion mutation PAK strain Pseudomonas aeruginosa

Project description:The bacterial type III secretion system (TTSS) is dedicated to directly effect host cell pathways by pathogens. In order to identify the key host responses of the various parts of the TTSS, we utilized expression profiling of a lung pneumocytes cell line, A549, exposed to various isogenic mutant strains of Pseudomonas aeruginosa PAK. We have devised a novel filtering method to isolate the key responses to the effector proteins as well as the translocation machinery. Individually, the effector proteins elicited host responses that were consistent with the known function of each, many of which either regulated, or are regulated by the cell cycle. However, our analysis has shown that the effector proteins elicit a distinct host expression pattern when present in combination, suggesting that these proteins function in synergy. Furthermore, the host transcriptional response to the translocation complex involved genes that are involved in remodeling of the plasma membrane, suggesting that the host cell is able to sense the protrusion of the TTSS machinery. This study shows that the individual components of the TTSS define an integrated system and that a systems biology approach is required to fully understand the complex interplay between pathogen and host. Comparison of the Pseudomonas aeruginosa PA2504 deficient strain and the PAO1161 wild type strain

Project description:Pseudomonas aeruginosa is an ubiquitous gram-negative bacterium that may colonize a wide range of organisms, including bacteria, plants, and animals. It is a human opportunistic pathogen which shows a great threat to immunocompromised patients. P. aeruginosa displays intrinsic resistance to many antibiotics, and has a high ability to develop novel mechanisms of resistance which forms a threat in hospital environments and makes it extremely hard to eradicate. Additionally over half of the genes of this bacteria have no described function, so it is urgent to search for proteins related to its pathogenicity and antibiotic resistance. The aim of this study was to characterise the P. aeruginosa PA2504 protein of unknown function. Basic phenotypic analysis did not indicate the role of PA2504 in the cell, thus, in order to recognize transcripts affected by the lack of PA2504 transcriptomes of the ΔPA2504 and the wild-type PAO1161 strains were compared using high-throughput RNA sequencing (RNA-seq). Using qRT-PCR method we determined that the level of PA2504 transcript is higher in the stationary phase of growth as compared to the exponential phase of bacterial growth (Log2 FC = 2,77) thus the samples for the RNA-seq experiments were withdrawn from this phase of growth.The RNA-seq revealed that the expression of 42 transcripts was changed in the ΔPA2504 mutant as compared to the parental PAO1161 strain and that the majority of them were connected to the sulphur transport/metabolism.
