Effect of ZYHT Recipe on transcriptome profiling of cancer cells

ABSTRACT: In order to explore the mechanism of ZYHT Recipe on gastric cancer, we performed a RNA-seq to explore the transcriptome profiling of gastric cancer cells treatment with or without ZYHT ,including three biological replicates per group.

Exploring the mechanism of Jiangtang Tiaozhi Recipe in the treatment of obese T2DM patients with dyslipidemia based on transcriptomics

Project description:Objective: To explore the mechanism of Jiangtang Tiaozhi Recipe in the treatment of obese T2DM patients with dyslipidemia based on transcriptomics. Methods: We chose 6 patients with obese type 2 diabetes mellitus and dyslipidemia (syndrome of excess of gastrointestinal heat) who were treated by JTTZR for 24 weeks, while 6 cases included in the healthy control group. We selected 6 cases in each group (disease group before treatment, disease group after treatment and healthy control group) to start the research of lncRNA microarray. According to the differentially expressions of lncRNAs and mRNAs, we secondly performed GO analysis, Pathway analysis, and found out some target lncRNAs as well as their associated mRNAs. The trial register number is NCT04623567. Results: (1) Disease group before treatment vs. healthy control group: There are 557 upregulated lncRNAs, 273 downregulated lncRNAs, 491 upregulated mRNAs and 1639 downregulated mRNAs. (2) Disease group after treatment vs. Disease group before treatment: There are 128 upregulated lncRNAs, 32 downregulated lncRNAs, 45 upregulated mRNAs and 140 downregulated mRNAs.

Quantitative proteomics revealed the mechanism of Yiqi Huoxue recipe used to treat cerebral infarction with Qi deficiency and blood stasis

Project description:Huoxue Yiqi (HXYQ) recipe is widely used to treat cerebral infarction with Qi deficiency and blood stasis syndrome (CI-QDBS), but its pharmacological mechanism is still not clear. In this study, the serum samples of patients with Qi deficiency and blood stasis syndrome in the convalescent stage of cerebral infarction were used as the research object. The protein expression levels among the control group, Yiqi group and Huoxue Yiqi group were compared by non labeled quantitative proteomics technology. The difference of protein expression level was higher than 1.2 times as the significant difference. Compared with the control group, there were 120 differentially expressed proteins; Through bioinformatics analysis and biological function clustering of differentially expressed proteins, it is found that Yiqi recipe can regulate energy metabolism and nerve function and change hemorheology, while Yiqi Huoxue recipe can regulate energy metabolism, antithrombotic function and improve vascular function. This study explored the biological mechanism of Yiqi recipe and Huoxue Yiqi recipe in the protein level, which provided a molecular basis for the clinical treatment of CI-QDBS and the related research of traditional Chinese medicine.

Label-free quantitative proteomics of gastric high-grade intraepithelial neoplasia

Project description:Purpose: Early detection and diagnosis are the keys to improving the survival rate and reducing the fatality rate linked to gastric cancer. The precancerous lesion of gastric cancer is referred to as Gastric high-grade intraepithelial neoplasia (HGIN). Both sensitivity and specificity of current biomarkers that aid in the diagnosis of gastric HGIN are still relatively low. The proteomic data of gastric HGIN are still scarce. The study aimed to explore candidate protein biomarkers for gastric HGIN screening with proteomics and bioinformatics technology. Methods: A total of 10 serum samples were collected and categorized into two groups, that is, the gastric HGIN and the healthy control groups. Label-free quantification (LFQ) in conjunction with liquid chromatography with tandem mass spectrometry (LC-MS/MS) was employed to identify the probable biomarkers for gastric HGIN. Further, differentially expressed proteins (DEPs) were quantified by the proteomic analysis. Results: In total, 1192 distinct serum proteins were discovered between the gastric HGIN group and the healthy control group. DEPs were recruited in the conduction of further analyses, utilizing a threshold of 1.5-fold difference in expression level (P ＜ 0.05) in comparison with the control group. There were 18 upregulated and 12 downregulated proteins in the gastric HGIN group in contrast with the control group. Bioinformatics analyses were performed according to Gene Ontology (GO), cluster, and KEGG pathway enrichment analyses. Conclusions: Our data provide more biological information for the formation of gastric HGIN and provide clues for further research on the pathogenesis of early gastric cancer.

Effect of synbindin on the gene expression profile of gastric cancer cells

Project description:In order to determine the effect of synbindin on the gene expression profile of gastric cancer cells, MGC-803 cells were infected with adenoviruses carrying either synbindin or GFP. The differentially expressed genes caused by synbindin ectopic expression suggests the underlying mechanism for synbindin-induced cell proliferation. Total RNA from each sample was amplified and transcribed into fluorescent cRNA using Agilent's Quick Amp Labeling protocol (version 5.7, Agilent Technologies). The labeled cRNAs were hybridized onto the Whole Human Genome Oligo Microarray (4 x 44K, Agilent Technologies). After washing, the arrays were scanned by the Agilent Scanner G2505C. 3 replicates per treatment (GFP adenovirus, synbindin adenovirus).

miRNA information of Yiqi-Bushen-Tiaozhi Recipe against NASH

Project description:MicroRNAs (miRNAs) have emerged as potential therapeutic targets for non-alcoholic fatty liver disease/non-alcoholic steatohepatitis (NAFLD/NASH). Traditional Chineses Medicine (TCM) plays an important role in the prevention or treatment of NAFLD/NASH. However, miRNA targets of TCM against NASH still remain largely unknown. Here, we showed that Yiqi-Bushen-Tiaozhi (YBT) recipe effectively attenuated diet-induced NASH in C57BL/6 mice. To identify the miRNA targets of YBT and understand the potential underlying mechanisms, we performed network pharmacology using miRNA and mRNA deep sequencing data combined with Ingenuity Pathway Analysis (IPA). Mmu-let-7a-5p, mmu-let-7b-5p, mmu-let-7g-3p and mmu-miR-106b-3p were screened as the main targets of YBT. Our results suggested that YBT might alleviate NASH by regulating the expression of these miRNAs that potentially modulate inflammation/immunity and oxidative stress. This study provides useful information for guiding future studies on the mechanism of YBT against NASH by regulating miRNAs.
