ISG induction differs immunogenical human splenic cDC2 activation from homeostatic ones

ABSTRACT: To assess the commonalities and differences in the transcriptomic reprogramming involved in immunogenic and homeostatic (DC2hm) human cDC2 activation, we conducted a comparison of gene regulation programs between DC2hm and immunogenically activated cDC2s (Generated using TLR-agonist cocktails or LPS+IFN-γ) by RNA-seq and found that though both programs upregulate common DC maturation markers like CCR7, CD80, CD40, and LAMP3, several transcription modules including ISG induction distinguish immunogenically activated cDC2s from DC2hm.

Project description:To test whether TSLP triggers the homeostatic maturation program of human splenic cDC2 (DC2hm program) in vitro, we administrated TSLP to in-culture human splenic cDC2 and performed RNA-seq. The results showed that TSLP induced DC2hm-like profile including the upregualtion of genes related to DC maturation, migration, immunoregulation, Treg induction and did not induce drastic ISG upregulation compared to either freshly isolated or cDC2s cultured in medium.

Project description:To explore the homeostatic maturation program of human cDCs, we exploited the nature of the spleen where DC maturation leads to migration from the red pulp and marginal zone to the white pulp of the spleen. Therefore, all steps of DC maturation are present and can be studied in the same organ. We first used single-cell RNA-seq (scRNA-seq) to profile lineage- (lin-) CD11c+ HLA-DR+ cells from spleens of 3 healthy human donors and identified a homeostatic maturation of the cDC2 subtype with 3 stages (cDC2, DC2pre-hm, DC2hm).

LAMP3-associated extracellular vesicles induce apoptosis in Sjögren’s disease

Project description:Sjögren’s disease (SjD) is an autoimmune disease that affects exocrine tissues and is characterized by increased apoptosis and decreased expression of membrane proteins involved in secretory function, such as Na-K-Cl cotransporter-1 (NKCC1), in salivary and lacrimal glands. Although the pathogenic mechanism triggering SjD is not well understood, overexpression of lysosome-associated membrane protein 3 (LAMP3) is associated with the disease in a subset of SjD patients and the development of SjD-like phenotype in mice. In this study, histological analysis of minor salivary glands of SjD patients suggested that LAMP3-containing material is being ejected from cells. Follow-on in vitro experiments with cells exposed to larger extracellular vesicles (LEVs) derived from LAMP3-overexpressing cells showed increased apoptosis and decreased NKCC1 expression. Proteomics identified LAMP3 as a major component of LEVs derived from LAMP3-overexpressing cells. Live-cell imaging visualized release and uptake of LAMP3-containing LEVs from LAMP3-overexpressing cells to naïve cells. Furthermore, experiments with recombinant LAMP3 protein alone or complexed with Xfect protein transfection reagent demonstrated that internalization of LAMP3 was required for apoptosis in a caspase-dependent pathway. Taken together, we identified a new role for extracellular LAMP3 in cell-to-cell communication via LEVs, which provides further support for targeting LAMP3 as a therapeutic approach in SjD.

We utilized the Chromium Single Cell Multiome ATAC + Gene Expression technique to depict the chromatin accessibility and nucleic mRNA profiles of lineage- (lin-) CD11c+ HLA-DR+ cells from the spleen of a healthy human donor and characterized a homeostatic maturation of the cDC2 subtype with 3 stages (cDC2, DC2pre-hm, DC2hm) at both transcription and epigenetic levels.

IRF4-regulated transcriptional heterogeneity of lung resident CD11b+ cDC2 subsets during influenza virus infection

Project description:Lung-resident conventional dendritic cells (cDCs) coordinate immune responses to respiratory viruses in the respiratory tract or after migration to mediastinal lymph nodes (mLN). Migratory DCs include cDC1s (CD103+XCR1+CD24hi) expressing IRF8 or cDC2s (CD11b+SIRPa+CD24+) expressing IRF4. IRF4+ cDC2s are divided into a CD24hi subset that requires IRF4 for differentiation and a CD24int subset that is present in the absence of IRF4. During influenza A virus (IAV) infection of mice, we characterized the kinetics of cDC2 subset accumulation in the lung and mLN and their differences in IRF4-dependent gene expression and function. We found that the two IRF4-expressing cDC2 subsets upregulated CD86 to high levels, produced IL-12p40 and the chemokines CCL17 and CCL2, and were capable of acquiring antigen in vivo and activating antigen-specific CD8+ T cells. Notably, the CD11b+CD24int cDC2 subset expressed canonical cDC markers and transcription factors and expanded to high numbers in the lung and mLN by day 6 post-infection. Transcriptome analyses on day 5 post-infection revealed that the CD11b+CD24int cDC2 subset expressed both IRF4 and IRF8 and harbored an elevated interferon response signature compared to the CD11b+CD24hi subset. Analyses of mice lacking Irf4 in CD11c+ cells showed that IRF4 promoted the function of CD11b+CD24int cDC2s, including the capacity to migrate to mLN and to produce CCL17 and CCL22, consistent with their altered gene expression profile in the absence of IRF4. In sum, our data show that the two lung-resident CD11b+ cDC2 subsets present in naïve mice elaborated distinct and common functional responses regulated by IRF4 during IAV infection.

Coordinated regulation and widespread cellular expression of interferon-stimulated genes (ISG) ISG-49, ISG-54, and ISG-56 in the central nervous system after infection with distinct viruses.

Project description:The interferon (IFN)-stimulated genes (ISGs) ISG-49, ISG-54, and ISG-56 are highly responsive to viral infection, yet the regulation and function of these genes in vivo are unknown. We examined the simultaneous regulation of these ISGs in the brains of mice during infection with either lymphocytic choriomeningitis virus (LCMV) or West Nile virus (WNV). Expression of the ISG-49 and ISG-56 genes increased significantly during LCMV infection, being widespread and localized predominantly to common as well as distinct neuronal populations. Expression of the ISG-54 gene also increased but to lower levels and with a more restricted distribution. Although expression of the ISG-49, ISG-54, and ISG-56 genes was increased in the brains of LCMV-infected STAT1 and STAT2 knockout (KO) mice, this was blunted, delayed, and restricted to the choroid plexus, meninges, and endothelium. ISG-56 protein was regulated in parallel with the corresponding RNA transcript in the brain during LCMV infection in wild-type and STAT KO mice. Similar changes in ISG-49, ISG-54, and ISG-56 RNA levels and ISG-56 protein levels were observed in the brains of wild-type mice following infection with WNV. Thus, the ISG-49, ISG-54, and ISG-56 genes are coordinately upregulated in the brain during LCMV and WNV infection; this upregulation, in the case of LCMV, was totally (neurons) or partially (non-neurons) dependent on the IFN-signaling molecules STAT1 and STAT2. These findings suggest a dominant role for the ISG-49, ISG-54, and ISG-56 genes in the host response to different viruses in the central nervous system, where, particularly in neurons, these genes may have nonredundant functions.

Project description:To explore whether co-expression of CCR7 and IL7R can better define DC2hm (homeostatically matured cDC2) than either marker alone. We sorted IL7R-CCR7+, IL7R+CCR7-, IL7R+CCR7+ DCs as well as CD141+ cDC1 and CD1c+ cDC2 from Lin-CD11c+HLA-DR+ splenocytes of 4 healthy donors and performed bulk RNA-seq. The results showed that the IL7R+CCR7+ double-positive subset showed the highest expression of DC2hm markers among the five subpopulations, supporting the hypothesis that combining CCR7 and IL7R better defines DC2hm.
