The combination of IL-7Ra and CCR7 is a highly specific marker to identify and purify homeostatically matured cDC2s (DC2hm).

ABSTRACT: To explore whether co-expression of CCR7 and IL7R can better define DC2hm (homeostatically matured cDC2) than either marker alone. We sorted IL7R-CCR7+, IL7R+CCR7-, IL7R+CCR7+ DCs as well as CD141+ cDC1 and CD1c+ cDC2 from Lin-CD11c+HLA-DR+ splenocytes of 4 healthy donors and performed bulk RNA-seq. The results showed that the IL7R+CCR7+ double-positive subset showed the highest expression of DC2hm markers among the five subpopulations, supporting the hypothesis that combining CCR7 and IL7R better defines DC2hm.

STAT5 inhibition abrogate TSLP-induced homeostatic maturation of human cDC2s

Project description:It has been reported that TSLP activates several downstream signalings including STAT5. To test whether TSLP triggers the homeostatic maturation program of human splenic cDC2 (DC2hm program) in a STAT5-dependant manner, we administrated TSLP to in-culture human splenic cDC2 with or without STAT5 inhibitor and performed RNA-seq. The results showed that TSLP-induced DC2hm-like profile was abrogated by STAT5 inhibition.

ISG induction differs immunogenical human splenic cDC2 activation from homeostatic ones

Project description:To assess the commonalities and differences in the transcriptomic reprogramming involved in immunogenic and homeostatic (DC2hm) human cDC2 activation, we conducted a comparison of gene regulation programs between DC2hm and immunogenically activated cDC2s (Generated using TLR-agonist cocktails or LPS+IFN-γ) by RNA-seq and found that though both programs upregulate common DC maturation markers like CCR7, CD80, CD40, and LAMP3, several transcription modules including ISG induction distinguish immunogenically activated cDC2s from DC2hm.

CITE-seq profiling of human splenic dendritic cells

Project description:To depict a comprehensive atlas of human splenic DCs and investigate DC2hm in a broad context, we performed Cellular Indexing of Transcriptomes and Epitopes by Sequencing (CITE-seq) using a panel of 151 oligo-conjugated antibodies recognizing surface proteins on Lin [CD3/CD19/NKp46]⁻CD66b⁻ CD88⁻CD89⁻HLA-DR⁺ splenocytes from two donors. This dataset identified nearly all currently recognized human splenic DC subsets. Integrated transcriptomic- and protein-level analyses support the conclusion that DC2hm represents a homeostatic maturation state originating from cDC2 and is distinct from other DC lineages.

Manifold roles of CCR7 chemokine GPCR in human trophoblast differentiation

Project description:CCR7 chemokine G protein-coupled receptor is expressed in extraembryonic tissues of the early human embryo, including trophectoderm and its derivatives cytotrophoblast (CTB), extravillous trophoblast (EVT), and syncytiotrophoblast (STB). However, its function in placental development remains poorly explored. Here, we generated human embryonic stem cells harboring CCR7 deletions and differentiated them into human trophoblast stem cell (hTSC) and hTSC-derived trophoblast organoids. We found that CCR7 mutant EVTs retained hTSC-like characteristics, exhibited decreased epithelial-to-mesenchymal transition, and reduced cell motility. Additionally, CCL21-CCR7 induced EVT terminal differentiation into endovascular EVT-like cells. Transcriptional profiling in CCR7 KO STBs identified reduced viral defense gene expression related to the protection against maternal-fetal transmission. Investigation of trophoblast organoids using single cell transcriptome profiling showed that CCR7 mutant trophoblast organoids comprised a smaller EVT population, but a larger STB population, compared to wild type organoids, while CellChat analysis indicated altered cell-cell communication including WNT, ACTIVIN, and IFN-I signaling pathways. Mechanistically, we found that CCR7 limited the cell-cell fusion of early STB differentiation by reducing cAMP levels. Together, our studies demonstrate that CCR7 plays multiple roles in cellular decision-making during trophoblast differentiation, promoting EVT differentiation, and limiting cell-cell fusion during early STB differentiation.

TCR repertoire of effector CCR7-CD38+ cells accumulating during experimental infection with Typhoidal Salmonella

Project description:In order to characterize pathogen specific T cell responses against Salmonella volunteers challenged with Salmonella enterica serovar Typhi (S. Typhi) or Salmonella Paratyphi A (S. Paratyphi). we used mass cytometry, to identify effector CD4+ T cells circulating during infection. We identified a population of CCR7-CD38+ cells accumulating during infection, and via unbiased single cell cloning and expansion we demonstrated that these CCR7-CD38+ cells are enriched in Salmonella specific T cells. In this experiment we performed TCR repertoire analysis of CCR7-CD38+ and CCR7-CD38- cells to determine the clonality of CCR7-CD38+ cells, the overlap between the repertoire of CCR7-CD38+ cells and of non-activated effector CCR7-CD38- cells, and to identify within CCR7-CD38+ and CCR7-CD38- cells the presence of the CDR3b TCR sequence of the pathogen specific T cell clones isolated from CCR7-CD38+ cells
