Assessment of the Anti-Tumor Effects of Poly(I:C) Combined with QS-21 on Melanoma Cells(B16F10)

ABSTRACT: This study explores the synergistic antitumor potential of Poly(I:C) and QS-21 in melanoma treatment. Through in vitro assays and animal models, we evaluated their combined impact on melanoma cell proliferation, apoptosis, and immune response. Our results indicate that Poly(I:C), an immunostimulant, activates the immune system to bolster antitumor activity. QS-21, an immunological adjuvant, enhances Poly(I:C)'s effects, together acting as an in situ vaccine to expose neoantigens and reignite the tumor-immune interaction. This research offers a promising immunotherapeutic approach for melanoma.

Project description:The goal of this study was to identify the transcriptional mechanisms involved in the activation of the immune system by QS-21, a triterpene glycoside purified from the bark of Quillaja saponaria which has adjuvant activity in vivo. Saponins represent a promising class of vaccine adjuvant. Together with the TLR4-ligand MPL, QS-21 is part of the Adjuvant System AS01, a key component of the Malaria and Zoster candidate vaccines that display demonstrated clinical efficacy. However, the mechanism of action of QS-21 in this liposomal formulation is poorly understood. Upon intra-muscular immunisation, we observed that QS-21 rapidly accumulated in CD169+ resident macrophages of the draining lymph node where it elicited a local innate immune response. Depletion of these cells abrogated QS-21-mediated innate cell recruitment to the lymph node, dendritic cell (DC) phenotypic maturation as well as the adjuvant effect on T cell and antibody responses to co-administered antigens. DCs rather than lymph node-resident macrophages were directly involved in T cell priming by QS-21 as revealed by the decrease in antigen-specific T cell response in Batf3−/− mice. Further analysis showed that the adjuvant effect of QS-21 depended on the integration of Caspase-1 and MyD88 pathways, at least in part through the local release of HMGB1. Taken together, this work unravels the key role of lymph node sentinel macrophage in controlling the adjuvant effect of a molecule proven to improve vaccine response in humans

ALFQ Adjuvanted HIV-1 Envelope Protein Vaccination Elicits Durable Functional Antibody and Cellular Responses in Nonhuman Primates

Project description:Adjuvants play an important role in modulating antigen-specific immune responses. We conducted a comparative adjuvant immunogenicity study in Rhesus macaques using HIV-1 subtype B gp120 envelope protein, B.63521 formulated with aluminum hydroxide gel (AH), or a family of liposomal adjuvants known as Army Liposome Formulation (ALF) to determine their effect on immune responses. ALF comprises saturated phospholipids, cholesterol, and an immunostimulant, monophosphoryl lipid A. Inclusion of a second immunostimulant, a saponin QS-21, or adsorption of the antigen to AH, followed by the addition of ALF generates ALFQ and ALFA, respectively, while the presence of both immunostimulants generates ALFQA. Priming with canarypox vector ALVAC, followed by boosting with ALVAC and gp120 formulated with AH, ALFA, ALFQ or ALFQA resulted in the QS-21-containing liposomal vaccine formulations (ALFQ and ALFQA) outperforming AH-based vaccine formulations (AH, ALFA) by inducing (i) binding antibodies of higher magnitude, durability, and avidity; (ii) higher neutralizing and Fc effector functional antibodies; (iii) higher frequency of durable antigen-specific plasma cells in the bone marrow; (iv) robust Env-specific polyfunctional CD4+ and CD8+ T cell responses and IL-21 producing T follicular helper cells. Transcriptomic analyses revealed that ALFQ and ALFQA vaccine formulations upregulated antiviral and innate immune pathways, including interferon signaling genes. These findings highlight that ALFQ is a highly potent adjuvant that facilitates multifaceted strong durable immune responses in nonhuman primates. Based on these data, an HIV vaccine containing B.63521 and CRF01_AE proteins formulated in ALFQ is currently being evaluated for safety and immunogenicity in a phase 1 clinical trial (NCT05423418).

Transcriptional profiling by RNA-seq of A. baumannii AB5075 wild-type, abaI::Tn26 and ∆abaM::Tn26 QS mutants.

Project description:Acinetobacter baumannii possesses a single divergent luxR/luxI-type quorum sensing (QS) locus named abaR/abaI. This locus also contains a third gene located between abaR and abaI which we term abaM that codes for an uncharacterized member of the RsaM protein family known to regulate N-acylhomoserine lactone (AHL) dependent QS and the expression of diverse genes in other β- and γ-proteobacteria. However, abaM has never been charatcerized in Acinetobacter baumannii, and the studies about the regulon of abaI are limited. In this study we revealed that AbaM differentially regulates at least 76 genes including the csu pilus operon and the acinetin 505 lipopeptide biosynthetic operon, that are involved in surface adherence, biofilm formation and virulence. A comparison of the wild type, abaM::Tn26 and abaI::Tn26 transcriptomes, indicates that AbaM regulates ~21% of the QS regulon including the csu operon. Moreover, the QS genes (abaI/abaR) were among the most upregulated in the abaM::Tn26 mutant. Overall, our data suggests that abaM is a key regulator of Acinetobacter baumannii QS and gene expression.

Microglial reprogramming enhances antitumor immunity and immunotherapy response in melanoma brain metastases

Project description:Melanoma is one of the tumor types with the highest risk of brain metastasis. However, the biology of melanoma brain metastasis and the contribution of the brain immune microenvironment to the responses to therapies remain insufficiently characterized. By using preclinical models and single-cell transcriptomics, we identify a mechanism to promote antitumor immunity in melanoma brain metastasis. We show that activation of the Rela/NF-kB pathway in microglia promotes melanoma brain metastasis and that targeting this pathway elicits microglia reprogramming towards a proinflammatory phenotype that enhances antitumor immunity and reduces brain metastatic burden. Additionally, proinflammatory microglial markers in melanoma brain metastasis correlate with better responses to immune checkpoint inhibitors in patients and we show that Rela/NF-kB targeting improves responses to these therapies in the brain. Thus, we propose targeting Rela/NF-kB in activated microglia as a strategy to promote antitumor immunity and responses to immune checkpoint inhibitors in melanoma brain metastasis.
