Expression comparison between SMC4 and conventional cultures

ABSTRACT: The undifferentiated state of pluripotent stem cells depends heavily on the culture conditions. We show that a unique combination of small molecules, SMC4, added to culture conditions converts primed pluripotent stem cells to a more naïve state. By conducting Affymetix analysis we show of majority of lineage markers are repressed in SMC4 culture.

Plasmodium condensin core subunits (SMC2/SMC4) mediate atypical mitosis and are essential for parasite proliferation and transmission

Project description:Condensin is a multi-subunit protein complex regulating chromosome condensation and segregation during cell division. In Plasmodium spp., the causative agent of malaria, cell division is atypical and the role of condensin is unclear. Here we examine the role of SMC2 and SMC4, the core subunits of condensin, during endomitosis in schizogony and endoreduplication in male gametogenesis. During early schizogony SMC2/SMC4 localize to a distinct focus, identified as the centromeres by NDC80 fluorescence and ChIP-seq analyses, but do not form condensin I or II complexes. In mature schizonts and during male gametogenesis there is a diffuse SMC2/SMC4 distribution on chromosomes and in the nucleus, and both condensin I and II complexes form at these stages. Knockdown of smc2 and smc4 gene expression revealed essential roles in parasite proliferation and transmission. The condensin core subunits (SMC2/SMC4) form different complexes and may have distinct functions at various stages of the parasite life cycle.

The proteomic study of serially passaged human skin fibroblast cells uncovers down-regulation of SMC4 involved in replicative senescence

Project description:In this study, a label-free quantitative proteomic approach was employed to analyze the serial passaged human skin fibroblast （CCD-1079Sk） cells, within 3305 proteins quantified. Of which, 372 proteins were significantly changed in early passage(P6), middle passage (P12) and later passage (P21), with a time-dependent decrease or increase tendency. Then, of which, SMC4 was selected for further biological validation. The results confirmed that the expression of SMC4 was significantly down-regulated in a time-dependent manner in the subculture of human skin fibroblasts (HSFb) with Western Blot experiment.

Induction of Ground-State Pluripotency by Minimal Factor Episomal-Expression in Single Cell Format

Project description:Human pluripotent stem cells in culture are often associated with the prime state which represents a more developed state relative to the naïve state which is often associated with the inner cell mass and thought to have the potential to give rise to all cell types. We have developed a small molecule-driven cocktail FMM that maintains human pluripotent stem cells in a state similar to the naive state as defined by several properties including gene expression profile. We used global gene expression to compare and contrast induced pluripotent stem cells -- from various starting points -- cultured in either small molecule medium FMM or conventional medium. Somatic cells were induced to reprogram by various methods and maintained in various culture conditions. Each culture condition was analyzed by various methods including global gene expression.

Secreted footprint of pluripotency in Man-13 HeSCs

Project description:With a view to developing a much-needed non-invasive method for monitoring the healthy pluripotent state of human stem cells in culture, we undertook proteomic analysis of the waste medium from cultured induced (Rebl.PAT) human pluripotent stem cells (hPSCs). Cells were grown in E8 medium to maintain pluripotency, and then transferred to FGF2 and TGFβ deficient E6 media for 48 hours to replicate an early, undirected dissolution of pluripotency. We identified a distinct proteomic footprint associated with early loss of pluripotency in both hPSC lines, and a strong correlation with changes in the transcriptome. We demonstrate that multiplexing of four E8- against four E6- enriched secretome biomarkers provides a robust, diagnostic metric for pluripotent state. These biomarkers were further confirmed by Western blotting which demonstrated consistent correlation with the pluripotent state across cell lines, and in response to a recovery assay.
