RNA-seq based in vivo transcriptomes of Korean fir (Abies koreana) with enhanced drought tolerance by Aureobasidium pullulans AK10

Project description:The conservation of the endangered Korean fir, Abies koreana, is of critical ecological importance. In our previous study, a yeast-like fungus identified as Aureobasidium pullulans AK10, was isolated and shown to enhance drought tolerance in A. koreana seedlings. In this study, the effectiveness of A. pullulans AK10 treatment in enhancing drought tolerance in A. koreana was confirmed. Furthermore, using transcriptome analysis, we compared A. koreana seedlings treated with A. pullulans AK10 to untreated controls under drought conditions to elucidate the molecular responses involved in increased drought tolerance.

Project description:The fullerenes, a kind of carbon nanoparticles, have potential for enhanced stress tolerance in plants. While the positive effects of polyhydroxy fullerene—fullerol on plants in response to drought at the physiological level have been documented, the molecular mechanism in Brassica napus are not entirely understood. In this study, exogenous fullerol was applied to the leaves of B. napus seedlings given drought. The leaves of B. napus seedlings in each treatment (sufficient water condition, drought, and drought combined with fullerol) were used to conduct the molecular mechanism using transcriptomic analysis.

The regulatory network of OsNAC6 targets drought-related genes orchestrating rice drought tolerance (RNA-Seq)

Project description:OsNAC6 is a stress responsive NAC transcription factor in rice known as a regulator for the transcriptional networks of the drought tolerance mechanisms. However, little is known about the associated molecular mechanisms for drought tolerance. Here, we identified OsNAC6-mediated root structural adaptation such as increased root number and root diameter that was sufficient to confer drought tolerance. Multiyear (5 years) drought field tests clearly demonstrated that OsNAC6 overexpression in roots produced higher grain yield under drought conditions. Genome-wide analyses revealed that OsNAC6 directly up-regulated 13 genes. Taken together, OsNAC6 is a valuable candidate for genetic engineering of drought-tolerant high-yielding crops.

Phosphoproteomic analyses the tetraploid Citrus wilsonii enhanced drought tolerance

Project description:In this study, 17 plants of tetraploid “Zhique” were firstly identified by screening 570 natural seedlings of Citrus wilsonii Tanaka. These tetraploid plants showed different morphology and exhibited significantly increased drought tolerance than the diploids via determination of leaf water potential, relative water content and electrolyte leakage. Large number of genes involved in photosynthesis-responsive were differentially expressed in tetraploids under drought stress by global transcriptome analysis, which was consistent with the detection of photosynthesis indicator including photosynthetic rate, stomatal conductance, chlorophyll and so on. Compared with diploids, phosphorylation modification also plays an important role in the tetraploids after drought stress through the transcriptional and protein level analysis. Additionally, the genes involved in the phenylpropanoid biosynthesis and starch and sucrose metabolism pathways were enriched in both tetraploids and diploids in response to water deficient. Importantly, tetraploids significantly take priority over the diploid via regulating plant hormone signal transduction, especially improving the levels of 3-indoleacetic acid, abscisic acid and salicylic acid and reducing gibberellic A3 and jasmonic acid contents. Collectively, our data reveals that synergistic regulation photosynthesis, phosphorylation modification and plant hormones accumulation contribute to drought tolerance of autotetraploid in Citrus wilsonii.

Expression data from ABA up-regulated rice under drought stress

Project description:To understand the molecular mechanism of drought stress resistance mediated by OsABA8ox3 gene, we checked the genome-wide expression profile changes in the OsABA8ox3 RNAi and WT seedlings using the Affymetrix GeneChip under the normal condition and drought stress. A total of 1436 genes showed greater than 2-fold higher expression levels in both WT and RNAi-9 seedlings after drought stress, and most of them had higher up-regulated folds in RNAi-9 seedlings than that of WT.
