Project description:Epidermal melanocytes form synaptic-like contacts with cutaneous nerve fibers but the functional outcome of this connection remains elusive. In this pilot study, we used our fully humanized re-innervated skin organ culture model to investigate whether melanocyte-nerve fibers interaction play a role in UV-B-induced melanogenesis. UV-B-irradiation significantly enhanced melanin content and tyrosinase activity in re-innervated skin ex vivo compared to non-innervated controls, indicating that neuronal presence is essential for exacerbating pigmentation upon UV-B irradiation in long-term culture. Comparative transcriptomic analysis between laser-capture-microdissected melanocytes from freshly embedded human skin and published microarray data on in vitro primary melanocytes identified Semaphorin-4A (SEMA4A) as possible mediator of melanocyte-nerve fibers interactions. SEMA4A protein levels in Gp100+ epidermal melanocytes were significantly higher in re-innervated skin than in skin alone, and reduced by UV-B treatment. Analysis of melanocytes in vitro showed reduced SEMA4A protein expression 24h after UV-B-irradiation while SEMA4A secretion into the medium was increased. In sensory neurons, conditioned media (CM) from UV-B irradiated melanocytes stimulated tubulin expression and axon growth. Re-transfer of CM from neurons that received CM from melanocytes back to melanocytes resulted into significant increased melanin content only when the CM derived from neurons previously receiving CM from UV-B irradiated melanocytes. These findings highlight the importance of melanocyte-neuron interactions for UV-B-induced melanogenesis and suggest that secreted proteins (e.g. SEMA4A) can function as a novel target in the treatment of hypo- and hyperpigmentation disorders.

Somatic mutations distinguish melanocyte subpopulations in human skin

Project description:To better understand the homeostatic mechanisms governing melanocytes, we performed deep phenotyping of single-cell expansions of melanocytes from human skin. In total, we interrogated the mutational landscapes, gene expression profiles, and morphological features of 297 melanocytes from 31 donors. To our surprise, a population of melanocytes with low mutation burden was maintained in sun damaged skin. These melanocytes were more stem-like, smaller, and less dendritic than their counterparts with high mutation burdens. We used single-cell spatial transcriptomics (10X Xenium) to reveal the spatial distribution of melanocytes inferred to have low and high mutation burdens (LowMut and HighMut cells), based on their gene expression profiles. LowMut melanocytes were found in hair follicles as well as in the interfollicular epidermis, whereas HighMut melanocytes resided almost exclusively in the interfollicular epidermis. We propose that melanocytes in the hair follicle occupy a privileged niche, protected from UV radiation, but periodically migrate out of the hair follicle to replenish the interfollicular epidermis after waves of photodamage. More broadly, our study illustrates the advantages of a cell atlas that includes mutational information, as cells can change their cellular states and positional coordinates over time, but mutations are like scars, providing a historical record of the homeostatic processes that were operative on each cell.

Project description:Mutational activation of the BRAF proto-oncogene in melanocytes reliably produces benign nevi (pigmented “moles”), yet the same change is the most common driver mutation in melanoma. One of the abiding mysteries of carcinogenesis is why oncogene activation doesn’t always lead to cancer. The reason nevi stop growing, and do not progress to melanoma, is widely attributed to a cell-autonomous process of “oncogene-induced senescence”. Senescence is usually accompanied by distinctive gene expression and various gene expression “signatures” have been developed to help investigators identify senescent cells and distinguish them from cells that have become growth arrested by other processes. Using a mouse model of Braf-driven nevus formation, we analyzed single-cell gene expression and found no evidence that nevus cells are senescent, either compared with other skin cells, or other melanocytes.

Comparison of gene expression signatures in human melanocytes after indirect cold physical plasma treatment and direct UV-irradiation

Project description:Cold atmospheric plasma is assembled out of various components such as UV-radiation, electrons, ions and an electromagnetic field at low temperature. In recent years cold atmospheric plasma gains interest as a medical tool used for improved healing of chronic wounds. In order to examine the impact of cold atmospheric plasma on the melanogenesis in human skin, the gene expression signature of differently treated lightly pigmented primary human melanocytes was analysed. Melanocytes were either treated in vitro indirectly by cold atmospheric plasma or irradiated by UV-radiation. After 2 h incubation the microarray analysis was performed.
