Myeloid Cell Replacement Therapy Enhances Brain Function in Friedreich Ataxia Mice via Intercellular Mitochondrial Transfer

ABSTRACT: We used single-cell RNA sequencing (scRNA-seq) to analyze the therapeutic effects of mitochondrial transfer by bone marrow-derived CD45+CD11b+ microglia-like cells (MGLCs) engrafted in the brain of FRDA mice. MGLCs were engrafted into the FRDA mouse brain following preconditioning with Busulfan and PLX3397 and total bone marrow transplantation. We then compared the gene expression of mKate2+ FA brain cells (mKate2+ cells) that received mitochondria from MGLCs and mKate2- cells (mKate2- cells) that did not.
