Circulating miRNAs in obese heart failure patients: Case control study

ABSTRACT: The main objective of this study was to identify differentially expressed circulating miRNAs in HF-obese (BMI ≥30 kg/m2 and % body fat > 25% in men and > 35% in women) and HF-lean patients (BMI < 25 kg/m2 and % body fat ≤22% in men and ≤32% in women). This case-control study was carried out using plasma samples from 20 HF patients (18-80 years with LVEF <45%) and from 10 healthy controls (blood donors) for microarray. The miRNAs were extracted with Trizol-LS reagent (396 μL plasma, 4 μL Cel-miR-39-3p, and 1.2 μL of Trizol-LS) and then analyzed on the miRNA 4.0 Affymetrix GeneChip array following the manufacturer's instructions. Raw data were normalized using Robust Microarray Average (RMA), batch effects were adjusted with Surrogate Variable Analysis (SVA), and differential expression analysis was performed with the Limma R package.
