Tuning BMP-Regulated Cell Differentiation in the Aortic Media by Mutating Matrix Gla Protein

ABSTRACT: The integrity of the vascular wall is crucial for maintaining the normal function of blood vessels. Matrix Gla protein (MGP), originally identified as an inhibitor of bone morphogenetic proteins (BMPs) and vascular calcification, is now recognized as a key vascular morphogen that guides cell differentiation across various tissues. In this study, we employed an MGP Pro64Gly knock-in mouse model alongside single-cell RNA sequencing to explore the role of MGP in mediating vascular cell differentiation. The Pro64Gly mutation specifically impairs MGP's ability to bind to and inhibit BMPs.
