Brain immune responses to systemic inflammation in postnatal mice


ABSTRACT: In newborn mouse, systemic bacterial infection damages the brain even in the absence of direct bacterial invasion. However, the early immune reactions by intracranial tissues remain an enigma. We injected lipopolysaccharide (LPS) intraperitoneally into 7-day-old mice to induce systemic inflammation. Brains were removed and dissociated 24 h after LPS injection, and CD11b(+) cells were isolated using the magnetic-activated cell sorting. Total RNA samples were extracted from the brain CD11b(+) cells and were analyzed with Gene Expression Microarray. The results indicated that the transcript expressions of the following nine genes were strikingly increased: Saa3, Saa1, Saa2, Cxcl13, Ccl5, Irg1, Cxcl9, Lcn2, and Slfn4. In situ hybridization indicated that Saa 3, 2, and 1, Cxcl13, Irg1, and Ccl5 were expressed by macrophages in the meningeal and perivascular space and choroid plexus, whereas Cxcl9 and Lcn2 were expressed by arachnoid, pial, and perivascular fibroblasts. In conclusion, immune reactions of the brain to systemic inflammation were initiated by leptomeningeal and choroid plexus macrophages and leptomeningeal and perivascular fibroblasts in postnatal mice.
