Molecular analysis of a microbial strain improvement paradigm

Project description:Industrial production of penicillins with the filamentous fungus Penicillium chrysogenum is based on an unprecedented effort in microbial strain improvement. Sequencing of the 32.19 Mb genome of P. chrysogenum Wisconsin54-1255 revealed many genes responsible for key steps in penicillin production. DNA microarrays were used to compare the transcriptomes of the sequenced strain and a penicillinG high-producing strain, grown in the presence and absence of the side-chain precursor phenylacetic acid. Transcription of genes involved in biosynthesis of valine, cysteine and alpha-aminoadipic acid, the amino-acid precursors for penicillin biosynthesis, as well as genes encoding microbody proteins, increased in the high-producing strain. Many key (intra)cellular transport processes involving penicillins and intermediates remain to be characterized at the molecular level. Genes predicted to encode transporters were strongly overrepresented among the genes transcriptionally upregulated under conditions that stimulate penicillinG production, illustrating potential for future genomics-driven functional analysis. Identification of the Streptomyces WblA regulon reveals drastic effects on development and specialised metabolism

Project description:Specialised metabolites made by the genus Streptomyces form the basis of 55% of clinically used antibiotics as well as cancer therapeutics, immunosuppressants and other anti-infectives. Streptomyces venezuelae is a model for the genus because it is fast-growing, sporulates in liquid culture within 24 hours and coordinates production of the antibiotic chloramphenicol with sporulation. In this work we demonstrate that the conserved orphan response regulator OrrA directly controls the production of the conserved WhiB-like protein WblA which in turn binds to 90 target gene promoters, including genes required for aerial hyphae production and sporulation. We provide evidence that WblA directly controls the production of AdpA and BldN, key developmental regulators that are required for entry into sporulation and show that the cellular levels of the key developmental regulators AdpA, BldM, BldN, RsbN, WhiA, WhiB and WhiD are significantly altered in a ∆wblA mutant. Proteins encoded by 32 of the 33 predicted specialised metabolite biosynthetic gene clusters in S. venezuelae are also significantly affected by loss of WblA and we show that the ∆wblA mutant over produces the antibiotic chloramphenicol. This is the first time the WblA regulon has been defined in the genus Streptomyces and is consistent with previous reports that deletion of wblA has pleiotropic effects on antibiotic production and sporulation in diverse Streptomyces species.

A new structural paradigm in copper resistance in a human pathogen

Project description:Copper is essential for both innate and adaptive immune function and copper resistance has emerged as an important determinant of virulence of microbial pathogens. In the human pathogen Streptococcus pneumoniae (Spn), cytoplasmic copper resistance is mediated by an operon encoding the copper-responsive repressor CopY, CupA, of unknown function, and CopA, a copper effluxing P1B-type ATPase. We show that CupA is a novel cell membrane-anchored Cu(I) chaperone for CopA, and that a Cu(I)-binding competent, membrane-localized CupA, like CopA, is obligatory for copper resistance. Bacteria were grown statically in Brain Heart Infusion media (Bacto BHI, Becton Dickinson) at 37C in an atmosphere of 5% CO2 to a culture density of OD620~0.2 and were processed as described in the related Sample record. Strain IU1781 (D39 rpsL1) served as the reference for the strain comparison. The experiment was repeated one time, and results are consistent with those observed by Shafeeq et al (Mol Microbiol. 2011). Data normalization was performed using the BioArray Software Environment (BASE 1.2; Saal et al, Genome Biol. 2002) using the Lowess (subgrid) method. Median spot intensities were normalized without background subtraction and used to calculate expression ratios. The cut-off for statistical significance of differential expression was set at an average up or down fold change of at least 1.8 fold.
