PolyA RNA seq of human glioblastoma cells treated with a pool of 11 miRNAs in vitro and in vivo (orthotopic tumors in mice)

ABSTRACT: Glioblastoma (GBM) has a poor prognosis due to infiltrative and therapy-resistant glioma stem-like cells. MicroRNAs (miRNAs) synergistically modulate biological pathways by repressing tumor-related genes, a mechanism lost in cancer. This study aimed to test a combination of different miRNAs exploiting their synergism as a multimodal therapy for GBM.

Project description:Glioblastoma cells infiltrate and incorporate into the normal brain parenchyma. We conduct unbiased multimodal single-cell and spatial sequencing to capture GBM cells and the surrounding tumor microenvironment using a regional sampling strategy. For each patient, we profile the peritumoral region, tumor edge, and tumor core, providing a comprehensive characterization of the transcriptional and epigenetic landscape of GBM spatial heterogeneity, with a specific focus on infiltrative cells.

A Zebrafish-Based Orthotopic Glioblastoma Xenograft Model for Preclinical Drug Screening and Clinical Drug Sensitivity Prediction

Project description:A reliable animal model that can mimic the GBM intracranial infiltration and Blood-brain barrier (BBB) interaction is necessary for effective therapeutics development. Here, we report a zebrafish-based orthotopic GBM xenograft model, in which GBM cells from different species and even patients, can robustly propagate and faithfully reproduce their histological characteristics. Single-cell RNA-seq indicates a transcriptomic adaption of GBM xenografts to infiltrative phenotype within the zebrafish brains. We also provide evidence that the BBB in zebrafish larva is molecularly and functionally intact and can interact with GBM cells in similar ways as in mammals, which together enables this model to accurately identify BBB penetrating drugs. Using GBM patients’ samples, we further generate zebrafish patient-derived orthotopic xenografts (z-PDOX) and proof-of-concept experiments indicate the short-term temozolomide response in z-PDOX can predict the long-term prognosis of corresponding GBM patients. These together illustrate the value of zebrafish GBM model in drug discovery and precision medicine.

Spatially resolved chimeric analysis reveals extracellular CKB-mediated ATP uptake at the glioma-astrocyte invasive front

Project description:Glioblastoma (GBM) is a highly invasive primary brain tumor that, although rarely metastasizing outside the brain, frequently infiltrates surrounding brain tissue. This infiltrative growth may exploit the brain’s metabolic support system, which involves various brain-resident cells providing essential nutrients and maintaining energy balance. In this study, we investigated the metabolic dependencies of infiltrative GBM cells by combining spatially resolved transcriptomic profiling with advanced computational methods in xenografted GBM models. Our findings reveal that GBM cells upregulate creatine kinase brain type (CKB) at the leading edge to adapt to local energy deprivation. CKB facilitates the conversion of creatine to phosphocreatine, utilizing ATP supplied by astrocytes to support tumor cell survival and invasion. Targeting CKB with cyclocreatine significantly reduced tumor size and restricted infiltrative growth in vivo, indicating that GBM cells utilize CKB to harness extracellular ATP via the creatine pathway. These findings suggest that targeting CKB in GBM may offer a novel therapeutic strategy to combat infiltrative growth in this aggressive cancer.

miRNA profiling of infiltrating glioma cells

Project description:Diffusely infiltrative growth of human astrocytic gliomas is one of the major obstacles to successful tumor therapy. Thorough insights into the miRNAs inolved in regulating glioma cell invasion thus appear of major relevance for the development of novel and advanced therapies. miRNA expression profiling on microdissected human tumor biopsy specimens was performed to identify differentially expressed miRNAs in invading glioblastoma cells in vivo. miRNA was extracted from the infiltrative rim (INF) as well as from the central tumor areas (CEN) of 7 patients with malignant glioma. The Micro Fluidic Card System (Applied Biosystems) was used according to the manufacturerM-bM-^@M-^Ys protocol for miRNA expression profiling.

Expression data of progesterone regulated miRNAs in U251 cells, derived from human glioblastoma

Project description:Glioblastoma multiforme (GBM), a grade IV astrocytoma, is the most common and aggressive brain tumor in adults, characterized by being highly infiltrative, angiogenic, and resistant to chemotherapy and radiotherapy. In previous works, has been determined that progesterone P4 increases proliferation, migration and invasion of cells derived from human GBMs through the interaction with its intracellular receptor (PR). In breast cancer, there exist evidence that P4 regulates the expression of miRNAs with tumor suppressor or oncogenic action, via the classical PR. The signature of miRNAs affected by P4 treatment in cells derived from human GBMs has not been determined. Therefore, we studied the effect of P4 on miRNAs expression pattern in U251 cells derived from a human GBM.

Glioma-initiating cells at tumor edge gain signals from tumor core cells to promote their malignancy

Project description:Intratumor spatial heterogeneity facilitates therapeutic resistance in glioblastoma (GBM). Nonetheless, understanding of GBM is limited to the resectable tumor core lesion while the seeds for recurrence reside in the unresectable tumor edge. In this study, stratification of GBM to core and edge demonstrated clinically relevant surgical sequelae. We establish regionally derived models of GBM edge and core that retain their spatial identity in a cell autonomous manner. Edge cells show a higher capacity for infiltrative growth, while core cells demonstrate greater therapy resistance. Investigation of intercellular signaling between these two cell populations uncovered the paracrine crosstalk from tumor core that provokes malignancy and therapy resistance of edge cells. These phenotypic alterations were initiated by HDAC1 in GBM core cells which subsequently affect edge cells by secreting the soluble form of CD109 protein. Our data reveal the role of intracellular communication between regionally different populations of GBM cells in tumor recurrence.
