Establishment and characterization of Bovine trophoblast stem cells (bTSCs) growing as adherent cells or vesicles

ABSTRACT: Understanding embryonic development still remains one of the main challenge in developmental biology. Using 3D in vitro models of early embryonic development in the mouse and human, scientists hope to decipher embryonic mechanisms, in particular the initial arrangement and interaction of cells within the embryonic structure. To date, there are very few models established for livestock species. In this study, we have derived and characterized bovine trophoblastic stem cells (bTSCs) that we further used to establish with bESCs (bovine embryonic stem cells) the first in vitro model of bovine blastoïds.

Establishing Bovine Embryonic Stem Cells and Analyzing Potential Mechanisms of their Self-Renewal

Project description:Bovine pluripotent stem cells (PSCs) hold significant potential for diverse applications in agriculture, reproductive biotechnology, and biomedical research. However, challenges persist in establishing stable bovine PSC lines and understanding the mechanisms un-derlying their pluripotency maintenance. Here, we derived bovine embryonic stem cells (bESCs) from Holstein cattle embryos. These cells exhibited robust differentiation capacity into three germ layers in vitro and in vivo. Transcriptome analysis revealed distinct molecular profiles compared to primed-state bESCs. Notably, bESC proliferation ceased on methanol-treated feeder cells, in contrast to mouse ESCs (mESCs), which proliferated normally. Pathway analysis identified key signaling events critical for bESC survival and proliferation, highlighting species-specific regulatory mechanisms. Furthermore, the derived bESCs demonstrated chimerism capacity in early bovine embryos, underscoring their functional pluripotency. This work provides a foundation for advancing bovine embryology research and stem cell-based biotechnologies in livestock.

Mixed-Lineage Leukemia 1 Inhibition Enhances the Differentiation Potential of Bovine Embryonic Stem Cells by Increasing H3K4 Mono-Methylation at Active Promoters. (RNA-seq)

Project description:Mixed-lineage leukemia 1 (MLL1) introduces 1-, 2- and 3-methylation into histone H3K4 through the evolutionarily conserved set domain. In this study, bovine embryonic stem cells (bESCs, known as bESCs-F7) were established from in vitro-fertilized (IVF) embryos via Wnt signaling inhibition; however, their contribution to the endoderm in vivo is limited. To improve the quality of bESCs, MM-102, an inhibitor of MLL1, was applied to the culture. The results showed that MLL1 inhibition along with GSK3 and MAP2K inhibition (3i) at the embryonic stage did not affect bESCs’ establishment and pluripotency. MLL1 inhibition improved the pluripotency and differentiation potential of bESCs via the up-regulation of stem cell signaling pathways such as PI3K-Akt and WNT. MLL1 inhibition decreased H3K4me1 modification at the promoters and altered the distribution of DNA methylation in bESCs. In summary, MLL1 inhibition gives bESCs better pluripotency, and its application may provide high-quality pluripotent stem cells for domestic animals.

Pyruvate and recombinant human basic fibroblast growth factor rescue the pluripotency and metabolism of bovine embryonic stem cells on low-density feeder layers

Project description:Feeder cells still play an important role in the in vitro culture of bovine embryonic stem cells(bESCs). A suitable microenvironment or niche is essential for the self-renewal and pluripotency of stem cells cultured in vitro. Feeder cells participate in the construction of stem cell niches by secreting growth factors and extracellular matrix proteins. In this study, we used metabolomics and transcriptomics to investigate the effects of feeder layer deficiency on bESCs. bESCs co-cultured with low-density feeder cells experienced a decrease in pluripotent gene expression, cell differentiation, and central carbon metabolic activity. When cell-permeable pyruvate (Pyr) and recombinant human basic fibroblast growth factor (rhbFGF) were added to the culture system, the pluripotency of bESCs on low-density feeder layers was rescued, and acetyl-coenzyme A(AcCoA) synthesis and fatty acid de novo synthesis increased. rhbFGF enhances the effects of Pyr and activates the overall metabolic level of bESCs grown on low-density feeder layers.

Efficient derivation of stable primed pluripotent embryonic stem cells from bovine blastocysts [ChIP-seq]

Project description:Embryonic stem cells (ESCs) are derived from the inner cell mass of preimplantation blastocysts. From agricultural and biomedical perspectives, the derivation of stable ESCs from domestic ungulates is important for genomic testing and selection, genome engineering, and modeling human diseases. Cattle are one of the most important domestic ungulates that are commonly used for food and bioreactors. To date, however, it remains a challenge to produce stable pluripotent bovine ESC lines. Employing a culture system containing fibroblast growth factor 2 and an inhibitor of the canonical Wnt-signaling pathway, we derived pluripotent bovine ESCs (bESCs) with stable morphology, transcriptome, karyotype, population-doubling time, pluripotency marker gene expression, and epigenetic features. Under this condition bESC lines were efficiently derived (100% in optimal conditions), were established quickly (3–4 wk), and were simple to propagate (by trypsin treatment). When used as donors for nuclear transfer, bESCs produced normal blastocyst rates, thereby opening the possibility for genomic selection, genome editing, and production of cattle with high genetic value.

Project description:Dendritic cell (DC)-based immunotherapy against glioblastoma multiforme is a novel treatment hope. Glioblastoma stem-like cells are, however, potentially causing immunoresistance. Glioblastoma cells cultured as gliomaspheres show a stem-like phenotype as opposed to classical adherent culture. They are thus a promising antigen source to specifically target glioblastoma stem-like cells via DC therapy and so overcome immunoresistance. Here we study the importance of gliomasphere-specific. Methodologically, we used 7 gliomaspheres, 3 of them patient-derived, as model system. Gliomasphere-specific protein expression was explored via quantitative proteomics.

Transcriptional profile of bovine chorioallantoic membrane explants in response to wild type, M-NM-^TvirB2 or M-NM-^TbtpB Brucella abortus

Project description:In this study the transcription profile of bovine trophoblastic cells infected with Brucella samples was evaluated. Chorioallantoic membrane (CAM) explants inoculated with wild type (strain 2308), and to compare with the transcription profile of trophoblastic cells infected with mutant strains lacking virB2 or btpB.M-NM-^TvirB2 or M-NM-^TbtpB by microarray analysis at 4 hours post infection. Genes with significant variation in levels of transcripts (fold change > 2 and P < 0.05) were functionally classified, and transcripts related to defense and inflammation were assessed by quantitative real time RT-PCR. Chorioallantoic membrane explant culture (CAM) were obtained from 7 intact pregnant bovine uteruses at the final third of gestation. Three-condition experiment, wild type B. abortus 2308, DvirB2 or DbtpB in 4 control replicates and Microarray analysis.
