FAM49B suppresses ovarian cancer cell growth through regulating MAPK signaling

ABSTRACT: The family with sequence similarity 49 member B (FAM49B, also known as CYRI-B) has recently attracted interest as a new player in cancer development. Upregulation of FAM49B expression has been observed in ovarian cancer tissues. Here we show that knockdown of FAM49B significantly increased ovarian cancer growth the in vitro, ovo and xenograft model. Transcriptomic analysis of FAM49B-depleted and control cells revealed that the MAPK pathway was upregulated upon FAM49B silencing. Supporting the transcription analysis, we found that knockdown of FAM49B enhanced the activation of EGFR and downstream MEK-ERK signaling, whereas FAM49B overexpression exhibited opposite trends. Overall, our data show that FAM49B acts as a suppressor of cancer cell growth in ovarian cancer through regulating MAPK signaling.
