Transcriptome of rNLS8 mice and controls injected with non-targeting control ASO

ABSTRACT: We asked how knockdown of genes associated with neurodegeneration using antisense oligonucleotides (ASO) affects disease phenotypes in an inducible TDP-43 transgenic mouse model (rNLS8, Walker et al., Acta Neuropathologica 2015) backcrossed to the C57BL/6 background. This dataset includes only the non-targeting control, which was also used as a replication cohort for transcriptional changes in rNLS8 mice (Rezaei et al., in revision at Nature Communications). We show cholesterol dysmetabolism and marker genes of disease-associated oligodendrocytes in the brain of rNLS8 mice. Data from mice receiving the targeting ASOs will be made available upon publication of the manuscript (Kocsis-Jutka et al, in preparation).

Bulk neocortical RNA sequencing of rNLS8 mice (on mixed C57BL/6J × C3H/HeJ background ) +/- active immunization with C-terminal hTDP-43 antigens

Project description:This data set reports RNA sequencing results obtained from whole cerebral cortices of NEFH-tTa/tetO-hTDP43∆NLS transgenic (“rNLS8”) mice that were actively immunized with C-terminal fragments of the human TDP-43 protein. The widely used rNLS8 mouse model overexpresses cytoplasmically mislocalized human TDP-43 in a doxycycline-inducible manner, mimicking ALS/FTD-like CNS pathology and motor dysfunction (Walker et al., 2015). The purpose of this study was to evaluate the safety profile and therapeutic potential of various human TDP-43 epitopes (up to approximately 40 amino acids in length) which were used as antigens for active immunization in a rapidly progressing mouse model of TDP-43 proteinopathy. To this end, we pre-screened 15 peptide antigens, collectively spanning the entire TDP-43 protein sequence, for immunogenicity and safety. Next, we repeatedly immunized “rNLS8” mice with the most promising antigens prior to transgene induction, and performed bulk RNA sequencing of the neocortex, since this region is considerably affected by TDP-43 pathology in both humans and the mouse model. We asked whether active immunization with two different peptide combinations leading to high antibody titers would affect – i.e. prevent – transcriptional alterations upon transgene induction. In summary, we provide a bulk neocortical gene expression profile of actively immunized (and mock-treated) ALS/FTD-resembling “rNLS8” mice.

Comparing the effects of live versus recorded relaxation on mood, coping, and quality of life on patients with cancer

Project description:Study hypothesis: 1. In patients attending the Oncology Health Centres for the first time, what is the relative effectiveness of live relaxation "LIVEREL" and recorded relaxation "RECREL" (i.e. "treatment as usual" including audio recorded relaxation as part of support within the Oncology Health Centres) on mood, coping and quality of life? 2. What is the relative cost-effectiveness of LIVEREL and RECREL? 3. What are the immediate effects of live versus recorded relaxation instructions on blood pressure and pulse rate? If patients randomised to LIVEREL have significantly higher relaxation scores on the Mood Rating Scale than those randomised to RECREL, we would conclude that the beneficial effects of LIVEREL was due to the extra social contact and/or the effects of live feedback on performance. Transcription profiling by array of Aedes aegypti infected with Wolbachia strains wMelPop and wMel

Project description:Wolbachia is a vertically transmitted intracellular bacteria that infect most than 60% of insect species. The strains wMelPop and wMel were introduced in the dengue virus vector Aedes aegypti, naturally not infected by Wolbachia. Recently, it was shown that those two strains inhibit dengue virus replication into their new host, A. aegypti (Moreira et al. 2009 and Walker et al. in preparation). The aim of this project is to look at the transcriptional response of Aedes aegypti to infection with wMel and wMelPop and try to find some genes or pathway potentially involved in the viral interference.Four laboratory lines of A. aegypti were used throughout this study. The PGYP1 and Mel2 lines were generated by transinfection with wMelPop and wMel strains respectively. PGYP1.tet and Mel2tet lines were treated with the antibiotic tetracycline and cured from Wolbachia infection (McMeniman et al., 2009 and Walker et al in preparation). The Mosquitoes were reared under standard laboratory conditions (26 ± 2 °C, 12:12 light/dark cycle, 75% relative humidity). Mosquito larvae were fed 0.1mg/larvae of TetraMin Tropical Tablets once a day. Adults were transferred to cages (measuring 30 x 30 x 30 cm) at emergence at 400 individuals per cage. Adults were supplied with a basic diet of 10% sucrose solution (Turley et al., 2009).
