Effect of adiponectin receptor agonist AdipoRON on breast cancer cell lines

ABSTRACT: Various adipocyte-derived adipokines reportedly regulate cancer growth, but their underlying mechanisms and cancer types suited for therapeutic interventions remain unknown. We analyzed adipokine receptor expression across cancer types and found that breast cancer cells express remarkably high levels of Adiponectin receptor 1. Adiponectin receptor stimulation decreased the proliferation and migratory ability of hormone receptor (HR)-positive and triple-negative breast cancer cell lines while activating apoptosis. Combination treatment with adiponectin receptor stimulants and existing drugs additively inhibited cell proliferation. Gene expression analysis revealed that adiponectin receptor stimulation regulates ferroptosis as well as ER expression in breast cancer cell lines. In conclusion, adiponectin receptor stimulation suppresses tumor growth across genetic breast cancer subtypes.
