The transcriptomic profiling of HCECs (PAI-2 knockout and control) exposed to PM2.5 for 3 hours

ABSTRACT: We previously found that PAI-2 mediates long-term PM2.5 exposure-induced corneal damage in vivo and in vitro, while its role during subclinical stage remains unclear. In this study,we constructed PAI-2 knockout-human corneal epithelial cells (KO) and the negative control (NC). Transcriptome analysis was performed in three groups: NC, PM2.5-exposed NC, KO and PM2.5-exposed KO. These analysis suggested cell-ECM interactions might play an important role in PAI-2-induced corneal injury upon PM2.5 exposure.

Transcriptomic Profiling of Human Corneal Epithelial Cells Exposed to Airborne Fine Particulate Matter (PM2.5)

Project description:Objectives: To explore the molecular mechanisms of PM2.5-induced dysfunction in human corneal epithelial cells (HCECs). Methods: RNA-Seq was performed to identify the differentially expressed genes (DEGs) in PM2.5-exposed HCECs compared to unexposed condition, which was followed by validation via real-time PCR (qRT-PCR). Results: A total of 434 DEGs—240 up-regulated and 194 down-regulated—were identified in PM2.5-exposed HCECs rather than unexposed HCECs. The expression of a few genes related to proliferation, inflammation, and aryl hydrocarbon stimulation were significantly altered by PM2.5 exposure, which may contribute to PM2.5-related corneal diseases. Conclusion: The present study identified the altered expression of hundreds of genes in HCECs exposed to PM2.5, which suggests the importance of PM2.5 for cornea health.

The effect of exposure to PM2.5 from the cowshed environment on the ovaries of rats

Project description:Fine particulate matter (PM2.5) is toxic to reproduction and can cause a range of reproductive disorders. However, the underlying mechanisms of female reproductive impairment due to cowshed PM2.5 exposure are unclear. The aim of this study was the investigation of the effects of PM2.5 on rat ovaries and its molecular mechanisms. Therefore, this study established a whole-body exposure model of PM2.5 in female rats to investigate the effects of PM2.5 on the ovaries. Exposure of rats to PM2.5 using a small animal whole-body PM2.5 exposure system. The device effectively simulates the exposure of animals or humans to PM2.5 within the barn environment, thereby offering enhanced scientific reference data. To put it briefly, for 30 days, the rats were exposed to six hours a day at four times the actual ambient PM2.5 concentration. Adult rats' respiratory coefficient, single-breath volume, and respiratory frequency were used to quantify their daily exposure to PM2.5. To study the effects of PM2.5 exposure on the ovaries of rats, the rats were divided into 2 groups: rats living in clean air were the Control group (Control), which received no additional treatments, and rats exposed to PM2.5 were the PM2.5-exposed group (PM2.5).

Fine particulate matter (PM2.5) induces microRNA-192–5p causing glomerular damage

Project description:To unravel changes in gene expression due to exposure to PM2.5, we performed bulk RNA-seq analyses of zebrafish larvae exposed to PM2.5 and controls. Among others, PM2.5 increased oxoglutarate alpha-ketoglutarate receptor 1a, nitric oxide synthase, arachidonate 5-lipoxygenase b, immunity-related GTPase family e1, sulfotransferase family 5A, and macrophage expressed 1. NADPH oxidase organizer 1a and NADPH oxidase 1 were upregulated due to PM2.5 exposure. Furthermore, protein tyrosine/serine/threonine phosphatase activity was decreased after exposure to PM2.5. GESA analysis showed that genes involved in proteasome complex formation, inflammatory and immune response, leucocyte-mediated cytotoxicity, peptidase activator activity protein folding, and apoptotic signaling were upregulated after exposure to PM2.5. These results indicate that PM2.5 exposure caused the activation of immune-inflammatory and oxidative stress pathways and lipid and metabolic dysregulation.

Esrrg inhibition protects against PM2.5-induced asthma aggravation by reducing Pde3b

Project description:PM2.5 exposure is closely linked to the exacerbation of asthma. Estrogen related receptor gamma (Esrrg), an orphan nuclear receptor, exerts a crucial role as a transcription factor in various metabolic diseases. Nevertheless, the impacts of Esrrg on PM2.5-triggered asthma aggravation have not been investigated. Herein, ovalbumin (OVA)-induced asthmatic mice were exposed to PM2.5 to establish a mouse model of asthma aggravation by PM2.5. In view of mRNA sequencing, Esrrg was the only member of nuclear receptor superfamily in the up-regulated differentially expressed genes in OVA compared with Naive groups as well as OVA+PM2.5 compared with OVA groups (|log2 (fold change)|>1 and p<0.05). In vivo, adeno-associated virus carrying Esrrg shRNA (AAV-shEsrrg) was applied to silencing Esrrg. In addition, Esrrg activity was suppressed pharmacologically with an inverse agonist GSK5182. Either AAV-shEsrrg or GSK5182 ameliorated airway inflammation in the PM2.5-aggravated asthmatic mice. In vitro, isolated mouse primary tracheobronchial epithelial cells (MTEC) from mice were identified by detecting cytokeratin 7-positive cells. The treatment of adenovirus vector with shEsrrg or GSK5182 mitigated the cell damage induced by PM2.5. Notably, phosphodiesterase 3B (Pde3b) expression was declined by Esrrg inhibition in vivo and in vitro. Dual luciferase reporter and ChIP-PCR assays showed the binding of Esrrg to the Pde3b promoter. Taken together, these results revealed that Esrrg inhibition alleviated airway inflammation in the PM2.5-deteriorated asthmatic mouse model and prevented PM2.5-driven MTEC injury through binding to the Pde3b promoter, which might contribute to further study the therapy of PM2.5-aggravated asthma.
