Project description:Isl1 encodes a member of the LIM/homeodomain family of transcription factors. During retinal development. Isl1 plays a critical role in RGC fate specification. Tbr2 is essential for the formation and maintaining the survival of ipRGCs. In this study, we tested whether ectopically expressing Isl1 in Tbr2-expressing cells affects the developmental program of Tbr2-expressing retinal neurons, including ipRGCs and a subset of wide-field displaced amacrine cells. We generated a Tbr2-Isl1 knock-in allele using gene targeting technique, and then used RNA-seq approach to compare transcriptome profiles between E15.5 wild-type (Tbr2-flox/+) and Tbr2-Isl1/+ retinas to uncover how Isl1 affects Tbr2-regulated genes.

ISL1 and POU4F1 directly interact to regulate the differentiation and survival of inner ear sensory neurons

Project description:The inner ear sensory neurons play a pivotal role in auditory processing and balance control. Though significant progresses have been made, the underlying mechanisms controlling the differentiation and survival of the inner ear sensory neurons remain largely unknown. During development, ISL1 and POU4F transcription factors are co-expressed and are required for terminal differentiation, pathfinding, axon outgrowth and the survival of neurons in the central and peripheral nervous systems. However, little is understood about their functional relationship and regulatory mechanism in neural development. Here, we have knocked out Isl1 or Pou4f1 or both in mice of both sexes. In the absence of Isl1, the differentiation of cochleovestibular ganglion (CVG) neurons is disturbed and with that Isl1-deficient CVG neurons display defects in migration and axon pathfinding. Compound deletion of Isl1 and Pou4f1 causes a delay in CVG differentiation and results in a more severe CVG defect with a loss of nearly all of spiral ganglion neurons (SGNs). Moreover, ISL1 and POU4F1 interact directly in developing CVG neurons and act cooperatively as well as independently in regulating the expression of unique sets of CVG-specific genes crucial for CVG development and survival by binding to the cis-regulatory elements including the promoters of Fgf10, Pou4f2, and Epha5 and enhancers of Eya1 and Ntng2 These findings demonstrate that Isl1 and Pou4f1 are indispensable for CVG development and maintenance by acting epistatically to regulate genes essential for CVG development.

Project description:Understanding factors that drive development and function of the sympathetic neuron is crucial to development of potential therapies for neuroblastoma. Here, we identify a key cell autonomous role for the LIM homeodomain transcription factor ISL1 for survival, proliferation and differentiation of sympathetic neurons throughout development. Analysis of several Isl1 mutant mouse lines, including one in which Isl1 was specifically ablated in sympathetic neuron (Wnt1-cre;Isl1 f/f) revealed an early requirement for Isl1 within sympathetic neurons for proliferation and surrvial. RNA-seq analyses on sympathetic neurons revealed dysregulation of a number of genes critical for sympathetic neuron development .Our studies demonstrated that ISL1 regulated the peoliferation, surrvial and differentiation.

Exploring the gene expression associated with Pten deficiency in the developing inner ear

Project description:In our recent study, we reported the function of phosphatase and tensin homolog (PTEN) during inner ear development. PTEN is necessary for neuronal maintenance, such as neuronal survival and accurate nerve innervations of hair cells. To better understand the genes and signaling networks related to auditory neuron maintenance, we examined the profiles of differentially expressed genes (DEGs) using microarray analysis in Pten-deficient mice at E14.5. We identified 46 statistically significant DEGs using Significant Analysis of Microarrays (SAM) analysis with a false discovery rate (FDR) equal to zero. Among the DEGs, expression levels of candidate genes and expression domains were validated by quantitative real-time polymerase chain reaction (RT-PCR) and in situ hybridization, respectively. Ingenuity pathway analysis (IPA) with DEGs identified significant signaling networks associated with cellular movement and axon guidance. Significant networks revealed that Spp1-mediated cellular movement and G-protein signaling 4 (RGS4)-Akt are related with axon guidance. This result was consistent with the phenotypic defects of spiral ganglion in Pten conditional knockout (cKO) mice (e.g., abnormal migration of spiral ganglion and irregular formation of neuritis). From this study, we suggest two key regulatory signaling networks mediated by Spp1 and RGS4, which may play potential roles in neuronal differentiation of developing auditory neurons.
