Salmonella typhimurium targets distal colonocytes to trigger infection [RNA-seq]

ABSTRACT: In order to ascertain the dynamic changes of transcriptomics throughout the whole cycle of Salmonella infection in the mouse intestine and offer basic support for the single-cell and spatial transcriptome data analysis in this research, we gavaged BALB/c mice with Salmonella and carried out transcriptome sequencing on the colon tissues at five time points during the entire infection cycle.

Project description:To quantify the extent of Salmonella infection in different large intestine segments, we performed spatial transcriptomics to analyse which clusters of cell populations were most infected by Salmonella and determined susceptibility in the distal colonocyte of mice. To further identify that Salmonella infection-associated genes and pathways were were both significantly expressed in distal intestinal segments following Salmonella infection, we used spatial transcriptomics data to delineate the different spots on a cell-by-cell basis for subsequent cellular annotation. Our analyses reveal the susceptibility of DCC upon Salmonella infestation.

Project description:To clarify how different intestinal cell types respond when facing the key intestinal pathogenic microorganism Salmonella, we conducted single-cell transcriptomic sequencing. The results revealed that a specific cluster of mature colonic epithelial cells was the most susceptible to infection. By describing the transcriptomic changes of this cluster of cells at different time points after infection, we further aimed to explain how this cluster of cells was affected by the infection.

Project description:We identified miRNAs differentially regulated upon Salmonella infection by comparative deep-sequencing analysis of cDNA libraries prepared from the small RNA population (10–29 nt) of HeLa cells infected with Salmonella (20 hpi) and mock-treated cells. Considering that at a MOI of 25 Salmonella is internalized in only 10-15% of the HeLa cells, we separated the fraction of cells which had internalized Salmonella (Salmonella+) from the bystander fraction (Salmonella-) by fluorescence-activated cell sorting (FACS), and extended the analysis of miRNA changes to these samples. Interestingly, we observed that Salmonella infection induces a significant decrease in the expression of all the detected members of the miR-15 family

Transcriptome analysis of Salmonella enterica serovar Typhimurium growth in physiological murine bile

Project description:Infection with Salmonella enterica serovar Typhi in humans causes the systemic, life-threatening disease typhoid fever. In the laboratory, typhoid fever can be modeled through the inoculation of susceptible mice with Salmonella enterica serovar Typhimurium. The ensuing disease is characterized by systemic dissemination and colonization of many organs, including the liver, spleen and gallbladder. Using this murine model, we previously characterized the interactions between Salmonella Typhimurium and host cells in the gallbladder and showed that this pathogen can successfully invade gallbladder epithelial cells and proliferate. Additionally, we showed that Salmonella Typhimurium can use bile phospholipids to grow at high rates. These abilities are likely important for quick colonization of the gallbladder during typhoid fever and further pathogen dissemination through fecal shedding. To further characterize the interactions between Salmonella and the gallbladder environment we compared the transcriptome of Salmonella cultures grown in LB or physiological murine bile. Our data showed that many genes involved in bacterial central metabolism are affected by bile, with the citric acid cycle being repressed and alternative respiratory systems being activated. Additionally, our study revealed a new aspect of Salmonella interactions with bile through the identification of phoP as a bile-responsive gene. Repression of phoP expression does not involve PhoPQ sensing of a bile component. Due to its critical role in Salmonella virulence, further studies in this area will likely reveal aspects of the interaction between Salmonella and bile that are relevant to disease.
